三氯乙烯用易致敏染毒法染毒SD大鼠所诱导的基因表达谱研究

摘  要: Trichloroethylene (TCE) is a volatile, colorless organic liquid, which is used mainly as a metal degreased solvent, an ingredient in adhesive, paint remover, typewriter correction fluid, and spot remover. As a result of the manufacture and disposal use in industrializedsociety,TCE is now an environmentalcontaminant in most underground water and surface water.Many adverse effects will arise when exposed to TCE. Skin damage (sclerosis and systemic dermatitis, which were associated with abnormal immune response),neurotoxicity, hepatotoxicity, nephrotoxicity, reproductive toxicity, fetal cardiac teratogenicity, auditory impairment, respiratory system toxicity, and cancer risk,which accompany with TCE exposure, have threatened the safety of lives on the earth.Previous explorations of the molecular and biochemical basis of TCE exposure mainly located on genes associated with carcinogenesis and affections of TCE metabolism and metabolites. However, the etiology and toxicological mechanism of many hazards brought by TCE exposure remained unknown. In this paper, experimental NIH mice and SD rats were selected to study theirresponses to TCE exposure after administration used to be adopted to induce allergicreaction. The purposes of this work were: (1) To find some evidences if TCE exposurewould induce abnormal immune response in animal;(2) To identify and characterizethe immune reaction related with TCE exposure;(3) To investigate the mechanisminvolved in immune disorder responses of TCE exposure.15 percent of TCE was injected intradermally on the rats backs (100 μ 1/120g) andmice backs (20 μ1/10g), in a interval of 7 days. Olive oil was injected on the backs ofanimals in control group by the same way. After 5 doses of application of TCE, thechallenged left-hind paws of SD rats were swollen significantly, while the TCEchallenged right ears of NIH mice showed no irritant response. NIH mice seemed lessresponsive than SD rats when exposed to TCE.Whole blood of SD rats was collected at 24h after the fifth or seventh intrader
