摘 要: 目的:研究唐草片对肝纤维化大鼠的保护作用并探讨其对NF-κB P65/IκBα信号通路的影响。方法:采用四氯化碳(CCl)联合猪血清的方法建立大鼠肝纤维化模型,72只SD大鼠按体重随机分为正常组、模型组、秋水仙碱组(0.12 mg/kg)、唐草片高中低剂量组(2,1,0.5 g/kg)。造摸第5周后,秋水仙碱组大鼠和唐草片组大鼠以体重按照剂量灌胃相应的药物,正常组大鼠和模型组大鼠灌胃同等容积的纯净水,持续8周后处死,取血和肝脏,苏木精-伊红和马松染色观察肝脏组织病理学改变;免疫组化染色法检测肝组织α-平滑肌肌动蛋白(α-SMA);全自动生化仪检测血清谷草转氨酶(AST)、谷丙转氨酶(ALT),Elisa法检测透明质酸(HA)、层黏连蛋白(LN)、Ⅲ型前胶原蛋白(PCⅢ)、Ⅳ型胶原蛋白(Ⅳ-C)、肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)水平;Western blot检测各组肝组织核转录因子(NF-κB P65)、核因子κB抑制蛋白(IκBα)。结果:与正常组比较,模型组大鼠肝组织出现明显肝纤维化,肝板结构排列紊乱,肝组织α-SMA,AST、ALT、HA、LN、PCⅢ、Ⅳ-C、TNF-α、IL-6表达水平显著升高(P<0.01);与模型组大鼠比较,唐草片组大鼠及秋水仙碱组大鼠可降低AST、ALT、HA、LN、PCⅢ、Ⅳ-C、TNF-α、IL-6的水平(P<0.05,P<0.01),肝组织病理学明显改善,同时可抑制NF-κB P65蛋白的表达,促进IκBα蛋白的表达。结论:唐草片对CCl4联合猪血清诱导的肝纤维化大鼠有保护作用,其作用机制可能与调控NF-κB P65/IκBα信号通路有关。 Objective:To investigate the protective effect of Tang herb on liver fibrosis in rats and the effect of Tang herb on NF-κB P65/IκBα signaling pathway.Methods:Carbon tetrachloride(CCl) combined with pig serum were used to establish rat liver fibrosis model. 72 SD rats were randomly divided into normal group, model group, colchicine group(0.12 mg/kg) and Tang herb high, medium and low dose group(2, 1, 0.5 g/kg) according to their body weight. The colchicine group and Tang herb group were intragastrically administrated with the corresponding drugs according to the dose of body weight, while the normal group and model group were intragastrically administrated with the same volume of pure water for 8 weeks.After the last drug administration, they were sacrificed, blood and liver samples were taken, Hematoxylin-eosin(HE) and Masson staining(Masson) was used to observe the liver histopathological changes;immunohistochemical staining(IHC) was used to detect the content of α-SMA in liver tissue;AST and ALT in serum were measured by automatic biochemical analyzer. The levels of hyaluronic acid(HA), laminin(LN), procollagen type Ⅲ(PCⅢ), collagen type Ⅳ(Ⅳ-C), tumor necrosis factor-α(TNF-α) and interleukin-6(IL-6) were determined by ELISA. Western blot was used to detect the expression of NF-κB P65 and IκBα protein in liver tissue. Results:Compared with the normal group, the liver tissue of the model group has obvious liver fibrosis with disordered liver plate structure, while α-SMA of liver tissue, AST、ALT、HA、LN、PCⅢ、Ⅳ-C、TNF-α、IL-6 levels increased significantly(P<0.01).Compared with the model group, the Tang herb group and the colchicine group could reduce the levels of AST、ALT、HA、LN、PCⅢ、Ⅳ-C、TNF-α、IL-6(P<0.05,P<0.01). The liver histopathology was obviously improved,and it could inhibit the expression of NF-κB P65 protein and promote the expression of IκBα protein.
