巴戟天多糖对大鼠骨髓间充质干细胞增殖和骨向分化的影响
Effects of polysaccharides from Morinda officinalis on proliferation and osteogenic differentiation of rat bone marrow mesenchymal stem cells

摘  要: 目的通过观察巴戟天多糖对骨髓间充质干细胞骨髓间质干细胞(BMSCs)增殖和骨向分化的影响。方法利用髓贴壁法分离培养BMSCs,并通过形态学、流式细胞仪检测细胞表面标志物以鉴定所取得的细胞;采用细胞计数(CCK-8)法检测不同浓度巴戟天多糖对BMSCs增殖情况的影响;对硝基苯磷酸盐法(p NPP)检测不同浓度巴戟天多糖对BMSCs碱性磷酸酶(ALP)活性的影响,以确定其最佳促分化的剂量。后续实验分空白组、巴戟天多糖组、成骨诱导组,在培养第7,14,21天分别收集各组细胞上清液,检测其ALP活性;培养第21天采用茜素红染色法检测各组BMSCs矿化情况。结果随着培养时间的延长,巴戟天多糖高、中浓度组可显著提高BMSCs的增殖(P<0.05),其中高浓度组效果最佳。与空白组相比,巴戟天多糖组可提高BMSCs上清液中ALP活性(P<0.05),增加其矿化结节数(P<0.05)。结论巴戟天多糖能促进BMSCs的增殖及向成骨细胞分化的能力。 Objective To observe the effects of Morinda officinalis polysaccharides on proliferation and bone differentiation of bone marrow mesenchymal stem cells(BMSCs).Methods BMSCs was isolated and cultured by marrow adherence method,the cell surface markers were detected by morphology and flow cytometry to identify the obtained cells.Cell counting kit-8(CCK-8)was used to detect the effects of different concentrations of Morinda officinalis polysaccharides on BMSCs proliferation.P-nitrophenyl phosphate(PNPP)method was used to detect the effects of different concentrations of Morinda officinalis polysaccharides on BMSCs alkaline phosphatase(ALP)activity in order to determine the optimal dose for differentiation.The follow-up experiments were divided into blank group,Morinda officinalis polysaccharide group and osteogenic induction group.The cell supernatants of each group were collected on the 7th,14th and 21st day of culture,and ALP activity was detected.The mineralization of BMSCs in each group was detected by alizarin red staining on the 21st day of culture.Results With the extension of culture time,the proliferation of BMSCs was significantly increased in the high and medium concentration groups(P<0.05),and the best effect was found in the high concentration group.Compared with the blank group,Morinda officinalis polysaccharide group increased ALP activity in BMSCs supernatant(P<0.05),and increased the number of mineralization nodules(P<0.05).Conclusion Morinda officinalis polysaccharides can promote the proliferation of BMSCs and its ability to differentiate into osteoblasts.
