千年健多糖提取工艺的优化及其生物活性研究

摘  要: 本研究优化了千年健多糖提取工艺,考察千年健多糖降血糖的活性。以提取温度、提取时间、料液比和醇沉浓度为影响因子,多糖得率为指标,寻找最优提取方案。通过考察对α-淀粉酶和α-D-葡萄糖苷酶的抑制活性,研究其降血糖活性。最佳提取参数:提取温度70℃,提取时间70 min,料液比1∶50,醇沉浓度80%,对α-淀粉酶和α-D-葡萄糖苷酶活性均具有较强的抑制作用,EC50分别为2.58、2.49 mg/mL。该工艺稳定可靠,千年健多糖具有较强的降血糖能力。 To optimize the extraction process of Homalomena occulta and investigate its biological activity,the effects of extraction temperature,extraction time,material-liquid ratio and alcohol precipitation volume fraction on the extraction rate of polysaccharides were investigated to find the optimal extraction scheme.By examining the inhibitory activity ofα-amylase andα-D-glucosidase,the hypoglycemic activity was investigated.The best extraction parameters were as follows:extraction temperature was 70℃,extraction time was 70 min,material-liquid ratio was 1∶50,alcohol precipitation concentration was 80%.Polysaccharides had a strong inhibitory effect on bothα-amylase andα-D-glucosidase activities,with EC50 of 2.58 and 2.49 mg/mL,respectively.The process was stable and reliable.Homalomena occulta polysaccharide had strong hypoglycemic capacity.
