摘 要: 目的建立一种新型的交叉引物等温扩增技术检测组织中的肠出血性大肠杆菌(EHEC),为临床诊断提供快速、灵敏、有效的检测方法。方法根据EHEC O157:H7的基因特征性保守序列,进行交叉、剥离引物设计之后,采用检测探针来进行O157:H7检测,从而检测出比较常见的食源性细菌并对这些细菌的特异性进行验证,对菌液进行不同浓度稀释,从而对交叉引物等温扩增技术检测的灵敏度进行评估,多次实验验证此方法的实用性。结果检测中,仅O157:H7菌株呈阳性,特异性良好,灵敏度也比较高。结论在EHEC O157:H7进行检测的过程中,应用交叉引物等温扩增技术来进行,操作简单易上手,检测速度比较快,检测灵敏结果准确,特异性比较强,应当广泛应用于肠出血性大肠杆菌O157:H7的检测实践当中。 Objective To establish a new cross-primer isothermal amplification technique for the detection of enterohemorrhagic escherichia coli(EHEC)in tissues,and to provide a rapid,sensitive and effective method for clinical diagnosis.Methods According to the conservative sequence of O157:H7 gene of EHEC,cross-primers and stripping primers were designed.Detection probes were used to detect O157:H7.The common food-borne bacteria were detected and their specificity was verified.The bacterial solution was diluted at different concentrations,thus the sensitivity of cross-primer isothermal amplification was improved.The practicability of this method is verified by many experiments.Results In the detection,only O157:H7 strain was positive with good specificity and sensitivity.Conclusion In the process of detection of EHEC O157:H7,cross-primer isothermal amplification technology is simple,easy to operate,fast,accurate and specific.It should be widely used in the detection of EHEC O157:H7.
