低剪切力下当归补血汤对内皮祖细胞功能损伤的保护作用

摘  要: 目的观察低剪切力(shear stress,SS)作用下当归补血汤对内皮祖细胞(endothelial progenitor cells,EPCs)功能的影响及调控机制。方法分离培养大鼠骨髓EPCs并鉴定,将EPCs随机分为6组:正常SS组和低SS组(M199培养液培养2 h)、辛伐他汀组(0.1μmol·L^-1的辛伐他汀培养2 h)、当归补血汤高、中、低浓度组(分别予以3.2 g·L^-1,1.6 g·L^-1,0.8g·L^-1的当归补血汤培养2 h),培养结束后弃上清,正常SS组加载1.2 Pa的正常SS,余各组加载0.12 Pa的低SS,分别在第30、60、360 min检测EPCs的增殖、迁移和成小管功能、一氧化氮(nitric oxide,NO)分泌、一氧化氮合酶(endothelial nitric oxide synthase,eNOS)mRNA及蛋白激酶B(protein kinase B,Akt)的表达。结果与正常SS组比较,低SS组EPCs功能随时间延长而受损,细胞分泌NO及表达eNOS mRNA、Akt蛋白在60min时迅速上调(P<0.01),随后在360 min时显著降低;与低SS组比较,辛伐他汀和当归补血汤高、中浓度可促进360 min时的细胞功能和NO分泌,上调eNOS mRNA及Akt蛋白的表达(P<0.05)。结论当归补血汤可通过调节NO/eNOS/Akt通路来保护低SS作用下受损的EPCs功能。 Objective To investigate the effect of Danggui Buxue Tang(DGBX)on the functional activity and the regulating pathway of endothelial progenitor cells(EPCs)exposed to low shear stress(SS).Methods EPCs isolated from bone marrow in rats were cultured and identitified,then were randomly divided into 6 groups:a normal SS group and a low SS group(preincubated with M199 medium for 2 h),a simvastatin group(preincubated with 0.1μmol·L^-1 simvastatin for 2 h),and 3 DGBX groups(preincubated with 3.2 g·L^-1,1.6 g·L^-1,0.8g·L^-1 DGBX for 2 h,respectively).Then the normal SS group was exposed to 1.2 Pa SS,the rest groups were all exposed to 0.12 SS.At each time point of 30 min,60 min,360 min,samples were collected for detection of the proliferation,migration,tubule formation,the secretion of nitric oxide(NO),the expression of endothelial nitric oxide synthase(eNOS)mRNA and protein kinase B(Akt),respectively.Results Compared with the normal SS group,the functional activities of EPCs in low SS group were declined with time;the secretion of NO,the expression of eNOS mRNA and Akt protein were all up-regulated obviously at 60 min(P<0.01),then down-regulated at 360min.The functional activities were improved in EPCs preincubated with simvastatin or DGBX(3.2 g·L-1,1.6 g·L-1)at 360min(P<0.05),as well as the NO/eNOS/Akt expression.Conclusion DGBX could protect the functional activity of EPCs exposed to low SS via the NO/eNOS/Akt pathway.
