摘 要: 目的:探讨岩藻甾醇对卵巢癌细胞分化、增殖及细胞周期的影响,并探讨其机制。方法:以HO8910、HRA及SKOV 3细胞株作为研究对象,选取对数生长期细胞,分为空白细胞对照、0.1 mmol/L岩藻甾醇组、0.2 mmol/L岩藻甾醇组及0.4 mmol/L岩藻甾醇组。观察各组细胞形态的变化;MTT检测转染岩藻甾醇对肿瘤细胞分裂增殖能力的影响;流式细胞仪检测并评价岩藻甾醇对肿瘤细胞分裂周期的影响;Western blot检测Prx III表达的变化。结果:岩藻甾醇可以诱导细胞以类圆形-星状-梭形的形态转变;与对照组相比,岩藻甾醇组细胞增殖率降低,G0/G1期细胞比例增加,S期细胞比例减少;与对照组相比,0.1 mmol/L、0.2 mmol/L及0.4 mmol/L岩藻甾醇组细胞中Prx III表达升高(F=18.053,P=0.000)。结论:岩藻甾醇可诱导卵巢癌细胞分化,抑制细胞增殖,其作用机制可能与调控Prx III的表达有关。 Objective:To investigate the effect offucosterol-on differentiation,proliferation and cell cycle of ovarian cancer cells,and to explore its mechanism.Methods:HO8910、HRA and SKOV 3 cells were divided into four groups:control group,0.1 mmol/L fucosterol group,0.2 mmol/L fucosterol group,0.4 mmol/L fucosterol group.Cell morphologic changes were observed under morphology.The effect of Adenanthin on the proliferation of tumor cells was detected by MTT assay.Cell apoptosis was detected by flow cytometry.The protein expression of Prx III was detected by Western blot.Results:Fucosterol could induce the morphological transformation of cells from circular-stellate-fusiform shape.Compared with the control group,the proliferation rate of algino sterol group decreased,the proportion of G0/G1 phase cells increased,and the proportion of S phase cells decreased.The expression of Prx III in the 0.1 mmol/L,0.2 mmol/L and 0.4 mmol/L algosterol groups were higher than those of the control group(F=18.053,P=0.000).Conclusion:Fucosterol can induce differentiation and inhibit ovarian cancer proliferation,and its mechanism may be related to Prx III expression.
