伏立诺他对急性重症胰腺炎大鼠早期炎性反应的影响

摘  要: 目的:探讨伏立诺他对急性重症胰腺炎大鼠早期炎性反应的影响。方法:选取健康Wistar大鼠24只作为研究对象,采用随机对照法将其分成Ⅰ组(空白对照组)、Ⅱ组(SAHA常规剂量组)、Ⅲ组(SAHA中等剂量组)、Ⅳ组(SAHA大剂量组),每组6只。观察四组血清淀粉酶、IL-6、TNF-a水平,胰腺组织NF-κB和p38 MAPK蛋白表达情况。结果:Ⅱ组、Ⅲ组、Ⅳ组血清淀粉酶、IL-6、TNF-a水平,胰腺组织NF-κB和p38 MAPK蛋白表达均高于Ⅰ组,随着SAHA剂量增加,血清淀粉酶、IL-6、TNF-a水平,胰腺组织NF-κB和p38 MAPK蛋白表达呈现逐步降低趋势,差异有统计学意义(P<0.05)。结论:伏立诺他可以降低急性重症胰腺炎大鼠早期炎性反应,利于重症胰腺炎的术后恢复,值得临床推广应用。 Objective To investigate the effect of suberoylanilide hydroxamic acid on early inflammatory response in rats with severe acute pancreatitis(SAP). Method 24 healthy Wistar rats as the research object,they were randomly divided into groupⅠ(control group),group Ⅱ(SAHA routine dose therapy group),group Ⅲ(SAHA medium dose group),group Ⅳ(SAHA high dose group),6 rats in each group. Serum amylase,IL-6 and TNF-α,pancreatic tissue NF-κB and p38 MAPK protein in pancreatic tissues were observed in four groups. Results Ⅱ,Ⅲ and Ⅳ group of serum amylase,IL-6 and TNF-α,pancreatic tissue NF-κB and p38 MAPK protein in pancreatic tissues were higher than that of group I,with the increase of SAHA dose,serum amylase,IL-6 and TNF-α,pancreatic tissue NF-κB and p38 MAPK protein in pancreatic tissues showed a trend of decrease gradually,P〈0. 05. Conclusion Suberoylanilide hydroxamic can reduce inflammatory reaction at early stage of severe acute pancreatitis in rats with severe acute pancreatitis,conducive to postoperative recovery,is worthy of clinical application.
