摘 要: 【目的】探讨加味四君子汤对慢性阻塞性肺疾病(COPD)小鼠膈肌线粒体损伤的修复作用及机制。【方法】将50只小鼠随机分为5组,即正常组,模型组,中药高、低剂量组和N-乙酰半胱氨酸(NAC)组,每组10只。除正常组,其余小鼠采用烟熏法复制COPD模型,持续8周。熏烟4周后,中药高、低剂量组给予加味四君子汤(剂量分别为20、10 g·kg-1·d-1)灌胃,NAC组给予NAC泡腾片混悬液(剂量为100 mg·kg-1·d-1)灌胃,正常组、模型组给予等体积生理盐水灌胃,共4周。给药结束后观察肺组织病理及膈肌线粒体结构变化,检测膈肌超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量及琥珀酸脱氢酶(SDH)活性变化。【结果】与正常组比较,模型组小鼠肺组织出现了符合COPD的病理改变,膈肌线粒体结构受损,而各给药组肺组织、膈肌病理表现较模型组有所改善。与正常组比较,模型组SOD、SDH活性显著降低(P <0.05),而MDA含量显著升高(P <0.05);与模型组比较,中药高、低剂量组SOD、SDH活性显著升高(P <0.05),MDA含量显著下降(P <0.05);与NAC组比较,中药高剂量组对SOD、SDH活性的升高作用及对MDA含量的降低作用更明显(P <0.05)。【结论】加味四君子汤可减轻COPD小鼠氧化应激水平,保护膈肌线粒体结构免于损伤,维持线粒体能量的正常代谢。 Objective To explore the protective effect of modified Sijunzi Decoction against diaphragmatic mitochondria injury in mice with chronic obstructive pulmonary disease and its mechanism. Methods Fifty mice were randomly divided into 5 groups, namely normal group, model group, high-and low-dose Chinese medicine groups and NAC group,10 mice in each group. All mice apart from the normal group were exposed to cigarette smoking for 8 weeks to induce into COPD model. After 4 weeks for exposure,the high-and low-dose Chinese medicine groups were given intragastric administration of modifiod Sijunzi Decoction(at the dosage of 20,10 g·kg-1·d-1,respectively),the NAC group was given intragastric adminstration of NAC suspension(at the dosage of 100 mg·kg-1·d-1),and the normal group and model group were given intragastric administration of the same volume of normal saline, the treatment lasting 4 weeks. The pulmonary histopathology and the structural changes of diaphragmatic mitochondria were observed after medication, and the superoxide dismutase(SOD)activity,malondialdehyde(MDA)content and succinate dehydrogenase(SDH)activity in diaphragmatic muscle were detected. Results Compared with the normal group,the model group showed pulmonary pathological changes conformed to COPD and the damaged structure of mitochondrias in the diaphragm. But the above changes in the model group were improved in the various medication groups. Compared with the normal group,the activities of SOD and SDH were decreased and MDA content was increased in the model group(P < 0.05);compared with model group,the activities of SOD and SDH were increased(P < 0.05),and MDA content was decreased(P <0.05) in the high-and low-dose Chinese medicine groups;compared with the NAC group, there were differences in the increasement of SOD,SDH and decreasement of MDA in high-dose Chinese medicine group(P < 0.05).
