摘 要: 目的建立丹参水提液中的成分数据库,阐明其中的成分,为研究丹参成分、寻找药物靶点提供科学依据。方法Agilent 1290-6550 Q-TOF-MS系统,色谱柱为ZORBAX ECLIPSE-C 18(4.6 mm×100 mm,3.5μm),流动相为0.1%甲酸水溶液(A)-甲醇(B)、梯度洗脱,流速为0.5 mL/min。经正、负离子全扫描模式搜集数据。结果经过对照品对照和文献检索,共指认了72个成分,其中包括26个酚酸类化合物、36个二萜类化合物、5个糖类化合物、3个有机酸类化合物和2个其他类化合物。结论超高压液相色谱串联四级杆飞行时间质谱技术(UPLC-Q-TOF-MS/MS)能快速、准确查找丹参水提液成分,为研究丹参谱效关系、筛选药物成分靶点提供新思路。 Objective The components database of Danshen water extract was established to clarify the ingredients,which would provide scientific basis for studying the components of Danshen and searching for drug target.Methods The system was carried out by a 1290 LC system in alliance with a 6550 Q-TOF-MS system,and ZORBAX ECLIPSE-C 18 column(4.6 mm×100 mm,3.5μm)was used.The mobile phase was made up of 0.1%formic acid(phase A)and methanol(phase B)at the flow rate of 0.5 mL/min,and the injection volume was 2μL.Data was collected by full scanning mode of positive and negative ions.Results 72 compounds were identified by a chemical profile of Danshen based on the experimental results as well as relevant articles,which consisted of 26 phenolic acid compounds,36 dicyandiamide,5 carbohydrate compounds,3 organic acid compounds and other kinds of compounds in total.Conclusion UPLC-Q-TOF-MS/MS can quickly and accurately find out the components of Danshen water extract,which provides a new way to study the spectrum effect relationship of Danshen and a new idea of screening drug target.
