摘 要: 建立了测定不同类别葡萄制品中白藜芦醇的高效液相色谱分析方法。根据样品基质不同,选用不同的前处理方法。采用Agilent C_(18)柱(4.6 mm×250 mm,5μm)进行分离,流动相为乙腈/水(体积比30∶70),流速为1.0 mL/min,柱温为35℃,检测波长为306 nm,外标法定量。白藜芦醇在0.50~50 mg/L范围内呈良好线性,线性相关系数为0.9999,加标回收率为91.3%~99.3%,相对标准偏差(RSD)为3.21%~5.67%,检出限为0.5 mg/kg。结果表明该方法适用范围广、重现性好和准确度高,可满足实验要求。 An analytical method for the determination of resveratrol in different grape products was established by high performance liquid chromatograph.Resveratrol was extracted by different pretreatment methods according to the sample matrix.The samples were separated on a Agilent C18 column(4.6 mm×250 mm,5μm).The mobile phase was acetonitrile/water(V/V,30∶70)at a flow rate of 1.0 mL/min and column temperature of 35℃.Detection wavelength was 306 nm.HPLC separation of resveratrol was quantified by the external standard method.The resveratrol had a good linearity in the range of 0.5~50 mg/L with a correlation coefficient of 0.9999.The recovery rate of samples were in the range of 91.3%~99.3%and the relative standard deviations(RSD)were in the range of 3.21%~5.67%.The detection limit was 0.5 mg/kg.The results show that the method has wide application range,good reproducibility,accuracy,and can meet the experimental requirements.
