HPLC结合化学计量学的茵陈提取物指纹图谱研究

摘  要: 目的:建立茵陈提取物的HPLC指纹图谱。方法:采用HPLC法,Agilent-C_(18)色谱柱(250 mm×4.6 mm,5μm);流动相为乙腈-0.2%甲酸溶液,梯度洗脱;流速1.0 mL/min;检测波长330 nm;柱温25℃;进样量10μL。采用相似度评价、聚类分析和主成分分析法对10批茵陈提取物进行系统归类和分析,并与茵陈药材进行对比。结果:确定了茵陈提取物中的10个共有峰,并对其中的9个峰进行了指认和鉴定,将不同来源的10批样品分为2类;茵陈提取物与茵陈药材的HPLC指纹图谱具有明显差异,表明在提取物制备过程中有机酸类成分可能发生水解或异构化反应。结论:所建立的茵陈提取物HPLC指纹图谱方法简便,重复性好,特征性强,可为茵陈提取物的质量控制提供实验依据。 Objective: The HPLC fingerprint of Artemisiae Scopariae Herba extract was established. Methods: The Artemisiae Scopariae Herba and Artemisiae Scopariae Herba extract were determinated based on the HPLC method. The chromatographic separation was performed on Agilent C(18) column( 250 mm×4. 6 mm,5 μm) with a mobile phase consisting of acetonitrile-0. 2% formic acid aqueous in gradient elution,at the flow rate of 1. 0 mL/min and the injection volume was 10 μL. The absorbance was monitored at 330 nm and the column temperature was 25 ℃. According to similarity evaluation,cluster method and principal component analysis( PCA),the Artemisiae Scopariae Herba extract collected from different sources compared with Artemisiae Scopariae Herba were classified and analyzed comprehensively. Results: The mutual mode of HPLC-UV fingerprints was set up,ten mutual peaks were marked,and nine peaks of them were identified based on HR-MS. Finally,ten bathes samples from different sources were divided into two groups. There are apparent differences of the HPLC fingerprint between Artemisiae Scopariae Herba and Artemisiae Scopariae Herba extract of extraction and preparation. The results showed that hydrolysis or isomerization of organic acids may occur during the preparation of extracts. Conclusion:This method is stable and reliable with a good reproducibility,which can provide a novel method for differentiation of Artemisiae Scopariae Herba extract.
