干姜五指方对脂溢性脱发小鼠毛发生长的影响

摘  要: 目的研究干姜五指方对脂溢性脱发模型小鼠毛发生长、激素水平和生长因子表达的影响。方法采用物理法小鼠背部脱毛,涂抹睾酮,建立脂溢性脱发模型。将90只C57BL/6J小鼠随机分为正常组,模型组,非那雄胺阳性对照组,干姜五指方高、低剂量组,并分别在脱毛区涂抹相对应的药物。观察小鼠毛发生长情况、毛囊形态及数目;检测皮肤组织睾酮(Testosterone,T)、二氢睾酮(Dihydrotestosterone,DHT)及雌二醇(Estradiol, E2)水平和血管内皮生长因子(Vascular endothelial growth factor, VEGF)、转化生长因子(Transforming growth factor,TGF)-β1的表达量。结果与模型组比较,干姜五指方高剂量组可显著促进小鼠毛发生长(P <0.01),增加小鼠毛囊数量(P <0.01),显著降低皮肤组织DHT水平(P <0.01);促进VEGF mRNA表达(P <0.05),并抑制TGF-β1 mRNA表达(P <0.05)。结论干姜五指方可显著促进脂溢性脱发模型小鼠毛发生长,可能通过抑制DHT水平,促进VEGF mRNA含量并抑制TGF-β1 mRNA表达发挥其药效。 Objective The effects of Ganjiang Wuzhi decoction on hair growth,hormone level and the expression of growth factor in C57 BL/6 J mice with seborrheic alopecia were studied.Methods Rosin/paraffin mixture was used for hair removal of mice and testosterone solution was then smeared to establish mouse model of Seborrheic alopecia.Ninety C57 BL/6 J mice were randomly divided into normal control group,model group,positive control group,high-and low-dose groups.Corresponding drugs were given separately.The morphological change and quantity of hair follicles were observed.Levels of testosterone(T),dihydrotestosterone(DHT)and estradiol(E2),and the expression of growth factors of skin tissue,including vascular endothelial growth factor(VEGF)and transforming growth factor(TGF)-β1 were determined.Results Compared with the model group,the high-dose of Ganjiang Wuzhi decoction can significantly accelerate hair growth on mice model with Seborrheic alopecia(P<0.01),increase the number of hair follicles in mice(P<0.01),decreased the level of the DHT in the skin tissues(P<0.01),promote the expression of VEGF mRNA(P<0.05),and inhibit the expression of the TGF-β1 mRNA(P<0.05).Conclusion Ganjiang Wuzhi decoction could promote the hair growth by inhibiting DHT levels,increasing the level of VEGF mRNA content and decreasing the expression of TGF-β1 mRNA.
