左旋肉碱和茶多酚对脂多糖诱导的小鼠急性肝损伤的保护作用及机制比较

摘  要: 目的从抗氧化应激和抗炎两方面探讨并比较左旋肉碱和茶多酚对脂多糖(LPS)诱导的小鼠急性肝损伤保护作用。方法40只雄性SPF级KM小鼠随机分为4组(n=10):假手术组(sham组)、模型组(LPS组)、左旋肉碱治疗组(LC+LPS组)和茶多酚治疗组(TP+LPS组)。腹腔注射LPS(10 mg/kg)方法复制小鼠急性肝损伤模型,sham组注射同样剂量生理盐水[1]。治疗组于术前7 d开始,持续8 d分别用左旋肉碱溶液灌胃(500 mg/kg,qd)[2]和茶多酚溶液灌胃(200 mg/kg,qd)[2-3]。造模24 h后眼球取血,分离血清用试剂盒检测血清中谷草转氨酶(AST)、谷丙转氨酶(ALT)、谷胱甘肽(GSH)、丙二醛(MDA)含量及超氧化物歧化酶(SOD)活力;手术摘取肝脏,并计算肝系数。用酶联免疫吸附实验(ELISA)检测肝组织匀浆液中肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)含量;用苏木精-伊红(HE)染色法显微镜下观察肝组织形态。结果与假手术组相比,模型组小鼠血清ALT、AST和MDA含量显著增加,血清中SOD、GSH含量下降,肝脏组织中炎性因子TNF-α、IL-1β含量增加,肝脏系数增加,镜下肝脏组织结构破坏严重。与模型组相比,两组治疗组上述指标均明显改善,且左旋肉碱缓解肝损伤效果优于茶多酚。结论左旋肉碱和茶多酚可通过抗氧化应激反应和抗炎性反应减轻LPS诱导的急性肝损伤且左旋肉碱缓解肝损伤效果优于茶多酚。 Objective To compare the protective effects of L-carnitine and Tea polyphenols on lipopolysaccharide-induced acute liver injury in mice,and to explore the mechanism from the perspectives of anti-oxidative stress and antiinflammatory.Methods A total of 40 male SPF grade KM mice were randomly divided into four groups:sham group(n=10),model group(LPS group,n=10),L-carnitine treatment group(LC+LPS group,n=10)and Tea polyphenol treatment group(TP+LPS group,n=10).The acute liver injury model was induced by intraperitoneal injection of LPS(10 mg/kg).From 7 days before LPS injection,the same dose of saline was injected into the sham group.The treatment group was given L-carnitine solution(500 mg/kg,qd)and Tea-polyphenol solution(200 mg/kg,qd),and lasted for 8 days.24 h after LPS or saline injection,blood was collected for testing the contents of AST,ALT,GSH,MDA and SOD in serum.Liver expression of tumor necrosis factor-α(TNF-α)and interleukin-1β(IL-1β)were measured by ELISA kits.HE staining and liver index were employed for determining the damage of liver tissues.Results Compared with the sham group,LPS injection significantly increased AST,ALT and MDA in serum;decreased SOD and GSH in serum;increased TNF-αand IL-βexpression in liver tissue which also showed typical tissue damage in HE staining slides.Compared with the LPS group,consecutive delivery of Lcarnitine and Tea-polyphenol significantly ameliorated the above indicators.Moreover,L-carnitine was better than Tea polyphenol in alleviating acute liver injury.Conclusion L-carnitine and Tea polyphenols could alleviate the acute liver injury induced by LPS through anti-oxidative stress and anti-inflammatory,and L-carnitine is better than Tea polyphenol in alleviating acute liver injury.
