摘　　要： 【目的】优化五达颗粒的最佳组方配比,为临床有效运用五达颗粒治疗腹部术后胃肠动力障碍提供可靠依据。【方法】将15只比格犬随机分为5组进行慢性实验。在胃窦、幽门、十二指肠、空肠和结肠浆膜植入应力传感器记录清醒犬胃肠运动。在胃体部安装1个不锈钢胃瘘管供灌入中药。术后1 d开始分别灌胃槟榔、人参、砂仁、乌药、桃仁颗粒剂混悬液,每味中药连续灌胃3 d。根据单味药物的检测结果进行五达颗粒组方配比优化。【结果】(1)槟榔5、10、20 g可剂量依赖性增强犬腹部术后全胃肠动力(P<0.05),且槟榔10 g和槟榔20 g作用差异不显著(P>0.05)。(2)人参9、18 g可增强犬腹部术后十二指肠、空肠和结肠动力(P<0.05)。(3)砂仁6、12 g可显著增加犬腹部术后胃窦和幽门振幅指数(P<0.05)。(4)乌药5、10、20 g未显著改变犬腹部术后胃肠收缩波数量、收缩振幅及振幅指数(P>0.05)。(5)桃仁10、20 g可增加犬腹部术后结肠振幅指数(P<0.05)。(6)在药典范围内,形成优化的五达颗粒配比(槟榔10 g、人参9 g、砂仁6 g、乌药10 g、桃仁10 g)可显著增强犬全胃肠运动(P<0.05),与其高剂量组方比较,差异均无统计学意义(P>0.05)。【结论】优化形成的五达颗粒含槟榔10 g、人参9 g、砂仁6 g、乌药10 g、桃仁10 g,具有促进全胃肠运动的作用,且在达到最大治疗效果时不增加各中药用量。 Objective To optimize the proportion of herbs in Wuda Granules to provide reliable basis for clinical effective application of Wuda Granules for the treatment of postoperative gastrointestinal motility disorder. Methods Fifteen beagles were randomly divided into 5 groups for the chronic experiment. Stress sensors were implanted into gastric antrum,pylorus,duodenum,jejunum and colon to record gastrointestinal movement in conscious beagles.A stainless steel gastric fistula was installed in the stomach body to perfuse Chinese medicine. From one day after the operation, the beagles in various groups were given intragastric administration of suspension of Chinese medicine Arecaesemen,Ginseng Radix et Rhizoma,Amomi Fructus,Linderae Radix,Persicae semen granules for 3 continuous days,respectively. And then according to the measure results of single drug,the proportion of herbs in Wuda Granules was optimized and compared. Results(1)Arecaesemen 5,10,and 20 g enhanced the postoperative whole gastrointestinal motility in dose-dependence manner, the difference in effects being insignificant between 10 g of Arecaesemen and 20 g of Arecaesemen.(2)Ginseng Radix et Rhizoma 9 and 18 g significantly enhanced the gastrointestinal motility of duodenum,jejunum and colon in beagles after operation.(3)Amomi Fructus 6 and 12 g significantly increased the amplitude index of gastric antrum and pylorus in beagles.(4)Linderae Radix 5,10 and 20 g did not significantly change the number of contraction wave,contraction amplitude and amplitude index in the stomach and intestine after abdominal operation.(5)Persicae semen 10 and 20 g significantly increased the colonic amplitude index after abdominal operation.(6)Accord with Pharmacopoeia,the Wuda Granules,with optimal ratio of Arecaesemen 10 g,Ginseng Radix et Rhizoma 9 g,Amomi Fructus 6 g,Linderae Radix 10 g,Persicae semen 10 g,significantly enhanced whole gastrointestinal motility of beagles after abdominal operation. And there was no significant difference compared with the high-dose formula
