银杏叶多酚物质不同提取方法的比较研究

摘  要: 【目的】观察不同提取方法对银杏叶中多酚成分含量的影响,为其进一步开发提供依据。【方法】采用乙醇提取法、酶解—乙醇提取法、超声波—乙醇提取法、微波—乙醇提取法制备银杏叶多酚成分,以称质量法测定提取物得率,福林—酚比色法测定多酚含量。【结果】应用不同提取方法的银杏叶多酚提取物得率及含量分别为:乙醇提取法(11.78±0.13)%、(5.42±0.06)%,酶解—乙醇提取法(12.71±0.09)%、(6.89±1.04)%,超声波—乙醇提取法(12.45±0.11)%、(6.71±0.06)%,微波—乙醇提取法(11.39±0.10)%、(5.97±1.00)%。经统计学分析,与乙醇提取法相比,酶解—乙醇提取法和超声波—乙醇提取法多酚提取物含量更高(P <0.05或P <0.01)。【结论】以上4种提取方法所得银杏叶多酚提取物的得率和含量不一,酶解—乙醇提取法和超声波—乙醇提取法更优。 Objective To observe the effects of different extraction methods on the contents of polyphenols from Folium Ginkgo, thus to provide evidence for further development. Methods The ethanol extraction method,cellulose enzyme-assisted extraction technology, ultrasonic-assisted extraction technology, and microwaveassisted extraction technology were adopted to prepare the polyphenols from Folium Ginkgo. The yield of polyphenos was determined by weighing method,and the content of polyphenols was measured by Folin-Ciocalteu colorimetry. Results The yields and contents of polyphenols extracted from Folium Ginkgo by different extraction methods were as follows respectively:the ethanol extraction method,(11.78 ± 0.13)%,(5.42 ± 0.06)%;the cellulose enzyme-assisted extraction technology,(12.71 ± 0.09)%,(6.89 ± 1.04)%;the ultrasonic-assisted extraction technology,(12.45 ± 0.11)%,(6.71 ± 0.06)%;the microwave-assisted extraction technology,(11.39 ± 0.10)%,(5.97±1.00)%. The results of statistical analysis showed that compared with the ethanol extraction method,the content of polyphenols extracted by the cellulose enzyme-assisted extraction technology and the ultrasonic-assisted extraction technology were higher(P < 0.05 or P < 0.01). Conclusion The yields and contents of polyphenols extracted from Folium Ginkgo by the above 4 extraction methods are different,and the cellulose enzyme-assisted extraction technology and the ultrasonic-assisted extraction technology can achieve higher yields and contents.
