AQP4抑制剂乙酰唑胺对癫痫持续状态大鼠血钾血钙的影响

摘  要: 目的观察AQP4抑制剂乙酰唑胺对癫痫持续状态大鼠血钾血钙的影响。方法将实验大鼠随机分为对照组、致痫组和乙酰唑胺干预组,观察行为学和脑电变化;检测各组大鼠血清K^+, Ca^(2+)浓度; HE染色观察海马CA1区神经元。结果行为学观察显示:对照组无明显癫痫发作;致痫组均出现Ⅳ~Ⅴ级癫痫发作,潜伏期短,持续时间长;乙酰唑胺干预组癫痫发作级别降低,潜伏期延长,发作持续时间缩短。脑电结果显示:对照组无痫样波,致痫组记录到棘波、尖波等痫样波,乙酰唑胺干预组痫样波减少。致痫组血清K^+浓度高于对照组, Ca^(2+)浓度低于对照组;乙酰唑胺干预组血清K^+浓度低于致痫组, Ca^(2+)浓度高于致痫组;组间差异有统计学意义(P <0.05)。HE染色显示,癫痫组海马锥体细胞核固缩,形态不规整;乙酰唑胺干预组锥体细胞排列紊乱明显改善。结论 AQP4抑制剂乙酰唑胺可抑制癫痫发作,其机制可能与下调血钾浓度、上调血钙浓度有关。 Objective To observe the effects of AQP4 inhibitor acetazolamide on serum potassium and serum calcium in rats with status epilepticus.Methods The rats were randomly divided into three groups:control group,epileptic group and acetazolamide intervention group,and the behavioral and electroencephalogram changes were observed;The concentrations of serum K+and Ca2+were measured in each group,and neuron morphology in hippocampal CA1 area was observed by HE staining.Results The behavioral observation showed that there was no obvious seizure in the control group;There were episodes of gradeⅣtoⅤ,with short incubation period and long duration in the epileptic group;The level of seizure was reduced,the incubation period was prolonged,and the duration of seizure was shortened in the acetazolamide intervention group.EEG showed no significant epileptic wave in the control group but spike wave and sharp wave in the epileptic group;The number of epileptic wave was conspicuously reduced in the acetazolamide intervention group.The serum K+concentration in the epileptic group was higher than that in the control group,and the Ca2+concentration was lower than that in the control group;The serum K+concentration in the acetazolamide intervention group was lower than that in the epileptic group,and the Ca2+concentration was higher than that in the epileptic group;The differences between the groups were statistically significant(P<0.05).The HE staining showed pyramidal nuclei pyknosis and irregular shape in hippocampus of epileptic group;Pyramidal cells arranged disorder was significantly improved in the acetazolamide intervention group.Conclusions The AQP4 inhibitor acetazolamide can inhibit seizures,and its mechanism may be related to down-regulating serum potassium concentration and up-regulating blood calcium concentration.
