摘 要: 目的研究中国寨卡病毒分离株的E基因多态性及其B细胞抗原表位的分布及变异情况。方法从NCBI中下载所有中国分离的寨卡病毒株、主要输入地委内瑞拉和萨摩亚的病毒株,从免疫表位数据库中检索获得寨卡病毒的B细胞抗原表位。使用MEGA7.0软件进行序列比对,分析中国寨卡分离株的基因变异情况及抗原表位的多态性。结果中国所有寨卡分离株均为亚洲型,E基因上有19个单核苷酸突变,其中3个异义突变,只有5个突变位点与来自委内瑞拉和萨摩亚的病毒株的突变位点相同。已验证的寨卡病毒B细胞抗原表位均存在于E蛋白上,尤其是DⅢ结构域。中国分离株和MR766株相比有3个B细胞表位发生改变,中国分离株自身产生的变异导致1个表位改变,B细胞表位的变异位点主要存在于DⅢ结构域。结论中国寨卡分离株总体变异率不高,与输入地病毒株相比也产生变异。有3个B细胞表位发生变异,DⅢ结构域是E蛋白上重要的抗原位点所在域且突变率高。 Objective To study the distribution and variation of B cell epitopes in the envelope of Zika virus in China.Methods The molecular data of Zika strains isolated in China,Venezuela and Samoa(from where most cases were imported into China)were extracted from NCBI database. B cell epitopes of Zika virus were retrieved from the immune epitope database(IEDB). Sequence alignment of MEGA 7.0 was used to analyze the genetic variation and polymorphism of antigenic epitopes. Results The genotype of all Zika virus isolated in China were Asian lineage. There were 19 single synonymous,only 5 mutations were identical to the mutation sites of the virus strains of Venezuela and Samoa. The verified B cell epitopes were all on the envelope protein,especially in the DⅢ domain. There were 3 B cell epitopes in Chinese isolates varied from MR766 strain;1 epitope was changed due to mutation of Chinese isolates. The variation sites of B cell epitopes mainly exist in the D Ⅲ domain.Conclusion Zika virus isolates of China were not highly varied in general,but they were varied from strains where they imported from,such as Venezuela and Samoa. Three B cell epitopes in envelope protein were mutated in China isolates. The DⅢ domain could be an important antigenic site and with high mutation rate.
