用于构建神经变性疾病模型的3D细胞培养研究进展

摘  要: 神经变性疾病是全球面临的重大疾病之一,传统的动物模型、2D细胞模型、脑片培养模型对理解疾病机制过程是有益的,但为了切实模拟患者大脑复杂神经网络,体现血脑屏障功能变化和炎症反应,基于3D细胞培养技术构建体外模型具有更大的潜能。本综述将介绍常见神经变性疾病现有模型、3D细胞培养体系、体外模拟这些疾病的障碍和当前在该领域的研究进展。 Neurodegenerative diseases are one of the major diseases in the world.The traditional animal model,2D cell model and brain slice culture model were the benefits for understanding the molecular mechanisms of disease.However,in order to effectively simulate the complex neural network of the patient s brain,reflecting the changes in blood-brain barrier function and inflammatory response,the 3D cell cultures should be utilized for its greater potential obtain in vivo cellular responses.In this review,we will focus on the progress of existing models of neurodegenerative diseases,3D cell culture systems,and current challenges or future directions to simulate neurodegenerative diseases in vitro.
