菟丝子水提物对黑色素合成的影响

摘  要: 目的研究菟丝子水提物对黑色素合成的影响。方法以斑马鱼胚胎为模型,检测不同浓度的菟丝子水提物对斑马鱼胚胎黑色素细胞形态、酪氨酸酶活性、黑色素含量的作用,及酪氨酸酶表达的情况。结果菟丝子水提物可以降低斑马鱼胚胎酪氨酸酶活性,减少黑色素的生成,作用强度与药物浓度呈依赖性,2mg/mL菟丝子水提物与50μg/mL熊果苷作用相似,但对酪氨酸酶蛋白表达并无明显影响。结论菟丝子水提物可以作用于斑马鱼胚胎黑色素细胞的酪氨酸酶,抑制黑色素合成。 Objective The present study aims to investigate the effect of water fraction from Se- men cuscutae (WFSC) on zebrafish larvae melanogenesis. Methods In accordance with traditional usage, the WFSC were extracted to deter-Results Conclusion mine the herbal effects on Zebrafish larvae by examining the activity of melanin contents, melanin morphology, tyrosinase activity in different concentration of WFSC. The melanocyte phenotypes of zebrafish larvae were observed as well. WFSC can reduce the zebrafish larvae tyrosinase activity, and decrease melanin formation. The degree of the effects is highly related to the level of the concentration,and the effect of 2mg/mL WFSC is similar to that 50g/mL arbutin but they had no obvious effect on tyrosinase protein ex- pression. WFSC significantly influences melanogenesis by regulating tyrosinase activity on Zebrafish larvae.
