维格列汀对大鼠非酒精性脂肪肝的影响

摘  要: 目的:探讨维格列汀对非酒精性脂肪肝(NAFLD)大鼠肝组织炎性因子表达的影响及其可能机制。方法:取SD大鼠24只随机分成3组,每组8只。模型组采用皮下注射CCL4溶液的方法建立NAFLD大鼠模型,正常组皮下注射等量生理盐水,干预组在建立NAFLD大鼠模型同时给予维格列汀灌胃。第8周断头处死大鼠,检测血清白细胞介素6(IL-6)、肿瘤坏死因子α(TNF-α)和丙氨酸转氨酶(ALT)、天门冬氨酸氨基转移酶(AST)水平;HE染色观察肝组织病理改变,实时PCR和免疫组化法检测肝组织解偶联蛋白2(UCP2) mRNA和蛋白表达。结果:干预组大鼠血清IL-6、TNF-α、ALT、AST水平较模型组均显著下降(均P<0.05)。病理检查显示,正常组肝小叶结构清晰完整,肝细胞未见明显脂肪变性;模型组肝细胞肿胀,胞质疏散,含脂肪粒,肝细胞核固缩;干预组肝细胞趋于正常,脂肪粒变小。模型组大鼠肝组织UCP2mRNA和蛋白表达均明显高于正常组和干预组(均P<0.05)。结论:维格列汀可抑制NAFLD大鼠肝组织UCP2表达,降低血清炎性因子水平,改善肝功能。 Objective:xmqqtgyx@qq.com To investigate the effect of vildagliptin on expression of inflammatory factors in liver tissue of rats with nonalcoholic fatty liver disease(NAFLD)and the possible underlying mechanism.Methods:A total of 24 SD rats were randomly divided into 3 groups(n=8 each).In the model group,a rat model of NAFLD was established by subcutaneous injection of CCL4 solution.The normal group was injected subcutaneously with the same amount of normal saline.The intervention group was given vildagliptin after modeling of NAFLD.The rats were sacrificed by decapitation after8 weeks,and the serum levels of interleukin-6(IL-6),tumor necrosis factor alpha(TNF-α),alanine aminotransferase(ALT),and aspartate aminotransferase(AST)were measured.HE staining was used to studythe pathological changes in the liver tissue;real-time PCR and immunohistochemistry were used to detect the mRNA and protein expression of uncoupling protein 2(UCP2)in the liver tissue.Results:The levels of serum IL-6,TNF-α,ALT and AST in the intervention group were significantly lower than those in the model group(all P<0.05).As revealed by pathology,the structure of hepatic lobules was clear and intact in the normal group with no obvious fatty degeneration in hepatocytes;the hepatocytes in the model group appeared swollen,with cytoplasmic evacuation,adipose granules,and nucleus pyknosis;the hepatocytes in intervention group tend to be normal with smaller adipose granules.The expression levels of UCP2 mRNA and protein in the liver of the model group were significantly higher than those in the normal group and the intervention group(all P<0.05).Conclusion:In NAFLD rats,vildagliptin may inhibit UCP2 expression in the liver tissue,reduce the level of serum inflammatory factors and improve liver function.
