离子色谱法-脉冲安培检测器测定奶粉中果聚糖的含量

摘  要: 建立离子色谱法分离检测奶粉中果聚糖的分析方法。果聚糖经过果聚糖酶溶液水解成果糖和葡萄糖,经离子色谱-脉冲安培检测器测定果糖的含量。采用CarboPac PA20色谱柱分离,以氢氧化钠-乙酸钠为流动相,梯度洗脱,使用脉冲安培检测器,建立了奶粉中果聚糖含量的分析方法。使用叠氮化钠及乙腈对实验前处理进行优化,结果表明试样中叠氮化钠浓度为10mg/L时为最佳。在0.8-16mg/L范围内的果糖呈良好的线性,相关系数均在0.999以上,对标准溶液进行6次平行测定的本方法的精密度小于1.0%。并进行了加标回收实验,奶粉中果聚糖的回收率在92.5-101%范围内。该方法具有操作简单、结果准确和快速有效等优点,可为奶粉中果聚糖的检测提供一个简单高效的方法。 An analytical method for the separation and determination of fructan in milk powder by Ion Chromatography was established.Fructose was hydrolyzed by fructanase solution to obtain sugar and glucose,and the content of fructose was determined by Ion Chromatography-pulse amperometric detector.Separation was carried out on a CarboPac PA20 column with sodium hydroxide-sodium acetate as the mobile phase,gradient elution,and pulse amperometric detector to establish a method for analyzing the content of fructan in milk powder.The pre-experimental treatment was optimized using sodium azide and acetonitrile.The results showed that the sodium azide concentration in the sample was 10 mg/L.The fructose in the range of 0.8-16 mg/L showed good linearity,and the correlation coefficient was above 0.999.The precision of the method for 6 parallel determinations of the standard solution was less than 1.0%.The spiked recovery experiment was carried out,and the recovery rate of fructan in milk powder was in the range of 92.5-101%.The method has the advantages of simple operation,accurate result,fast and effective,and can provide a simple and efficient method for detecting fructan in milk powder.
