微量全血单细胞凝胶电泳评定过度训练大鼠淋巴细胞DNA损伤

摘  要: 背景:微量全血单细胞凝胶电泳技术是检测有核细胞DNA损伤与修复的一种方法,具有敏感性高、取材方便、细胞制备简单、不需体外活化、费用低、实验周期短等优点。目的:采用微量令血单细胞凝胶电泳技术评定过度训练对大鼠血液淋巴细胞DNA损伤的情况。设计、时间及地点:随机分组设计、动物对照实验,于2004-08/09在沈阳体育学院研究生实验室完成。材料:雄性Wistar大鼠29只,按随机数字表法分为2组,正常对照组12只、运动组17只。方法:正常对照组大鼠不参加运动。运动组大鼠在玻璃池中进行游泳训练。采用6周递增负荷游泳训练建立过度训练动物模型,训练结束后24h,避光眼眶取血。主要观察指标:采用微量全血单细胞凝胶电泳法柃测单个血液淋巴细胞的DNA尾长、尾矩、椭圆矩,以反映DNA的损伤情况。结果:荧光显微镜下运动组大鼠淋巴细胞的DNA断裂比正常对照组明显,断片离开头部向阳极方向迁移,形成一个像彗星样的拖尾。运动组反映大鼠血液淋巴细胞DNA损伤的指标尾长、尾矩和椭圆矩显著高于正常对照组,差异有显著性意义(P〈0.01)。结论:6周递增负荷过度训练后大鼠血液淋巴细胞存在DNA损伤。 BACKGROUND: Micro-whole blood single cell gel electrophoresis assay is used to detect karyocyte DNA injury and repair. This method is characterized by high sensitivity, convenience, simple preparation, in vivo activation, low cost and short cycle. OBJECTIVE: To assess the effects of overtraining on DNA harms of lymphocytes using micro-whole blood single cell gel electrophoresis assay. DESIGN, TIME AND SETTING: The randomized control animal experiment was performed at the Graduate Student Lab in Shenyang Physical Education Institute from August to September 2004. MATERIALS: Twenty-nine male Wistar rats were randomly divided into 2 groups, a normal control group (n=12) and an exercise group (n= 17). METHODS: Rats in the normal control group did not do exercises. Rats in the exercise group swam in a glass pool. An incremental swimming exercise was adopted for 6 weeks to establish overtraining animal models. The rats were sacrificed and sampled after 24 hours in the condition of lightproof. MAIN OUTCOME MEASURES: DNA damage of lymphocytes of rats was measured by the method of micro-whole blood specimen in single cell gel electrophoresis, including DNA tail length, tail moment, olive moment. RESULTS: DNA fragmentation was significant in the exercise group than in the normal control group under a fluorescence microscope. Fragments moved towards positive pole, forming a tailing like comet. Tail length, tail moment and olive moment of lymphocyte DNA significantly increased in the exercise group than in the normal control group (P 〈 0.01). CONCLUSION: After having an incremental swimming exercise for 6 weeks, the DNA of lymphocytes was damaged.
