摘 要: [目的]克隆一个受CTV(CitrusTristezaVirus,柑橘速衰病毒)诱导的GAST1基因,通过生物信息学分析手段揭示其可能的作用方式。[方法]以克里曼丁橘cDNA为材料,通过扩增获得该基因,并利用生物信息学软件进行分析。[结果]该基因编码序列(CDS)长度为357bp,预测可编码含有118个氨基酸残基的蛋白质,其中半胱氨酸、脯氨酸和丝氨酸含量较高,分别占总蛋白的10.2%、8.5%和8.5%。生物信息学分析结果显示,CcGAST1编码的蛋白为具有信号肽的亲水性蛋白,具有GAST家族蛋白所特有的GASA结构域。多重序列比对结果显示和其他物种GAST1蛋白一样,CcGAST1蛋白C端非常保守,含有12个保守的半胱氨酸。以不同植物GAST1蛋白序列构建进化树,发现克里曼丁橘与甜橙亲缘关系最近,其次是核桃。[结论]CcGAST1富含半胱氨酸和脯氨酸残基,其保守的GASA结构域和12个保守的半胱氨酸残基可能与其功能相关。 [Objective] The aim of this study was to clone a CTV induced citrus GAST1 gene and to reveal its possible function mode by using bioinformatics analysis. [Method[ The gene was cloned from Citrus clementine by using PCR and was subjected to bioinformatics analysis. [Result] The length of its coding sequence (CDS) was 357 bp, which was predicted to encode a protein of 118 amino acids. CcGAST1 protein was rich of cysteine, proline and serine, which respectively account for 10.2%, 8.5% and 8.5%. Bioinformatic analysis result showed that the protein encoded by CeGAST1 was a hydrophilic protein with signal peptide and GASA domain. Multiple alignment analysis showed that, like other GAST1 proteins, CcGAST1 owns the GAST1 protein specific GASA domain and contains 12 conserved cysteines in the C terminal. According to the GASTl-based phylogenetic analysis, the closest relationship was found between Citrus clementine and Citrus sinensis, and the relationship between Citrus clementine and Juglans regia was also very close. [Conclusion] CcGAST1 is rich of cysteine and proline residues. The conserved GASA domain and the 12 cysteines in CeGAST1 may be closely bound up with its function.
