1,25(OH)_2D_3在少突胶质细胞慢性缺氧中的作用

摘  要: 目的观察1,25(OH)_2D_3在少突胶质细胞慢性缺氧模型中的作用,为血管性痴呆的治疗提供理论基础。方法提取新生SD鼠皮质混合胶质细胞进行原代细胞培养,震荡分离后通过增殖及诱导分化得到成熟的少突胶质细胞。并且应用荧光免疫标记进行鉴定。随后在其中加入非致死量的氯化钴制造少突胶质细胞慢性缺氧模型。随后将其分为3组:空白对照组A、实验组B、给药对照组C。B、C两组分别给予1,25(OH)_2D_3以及孕激素。并分别设定3个时间点。分别观察三组少突胶质细胞的形态,应用MTT确定细胞存活率,应用Westen blot确定Akt相关抗凋亡蛋白的表达。结果给予1,25(OH)_2D_3的实验组B的细胞形态以及细胞存活率明显高于空白对照组A以及孕激素对照组C。并且与对照组A、C相比,实验组B抗凋亡蛋白的表达上调。结论 1,25(OH)_2D_3能够在少突胶质细胞慢性缺氧模型中起到保护作用,并且上调抗凋亡蛋白的表达。 Objective To observe the effect of vitamin D on a chronic hypoxia model of oligo- dendroglia in rats and to find a new way for the treatment of vascular dementia. Methods The cortex less glial progenitor cells of newborn SD rat in primary cell culture were extracted, then the cells were purified by shock separation and the cells were obtained by proliferation and dif- ferentiation of oligodendrocytes. The application of fluorescent immune markers were utilized to identify the cells. A lethal dose of cobalt chloride was added to manufacture chronic hypoxia model of cell. Then the model was divided into three groups : Control group ( A), experimental group (B) and give medicine control group (C). Group B and C were given 1,25(OH)ED3 and progesterone respectively. The condition of the oligodendrocytes was observed, the cell sur- vival rate was determined by MTT at 12 hours, 24 hours and 48 hours. The expression of AKT related of antiapoptotic genes were detected by Western blot. Results The cells of group B were much better in cell morphology. And the cell survival rate of group B was higher than the blank control group A and progesterone control group C. The expression of antiapoptotic protein of B group was accelerated than that of the other two control groups. Conclusion 1,25(OH) 2D3 can protect oligodendrocytes chronic hypoxia model, proteins expression.
