维甲酸对支气管哮喘患者外周血Th17/Treg细胞功能的影响

摘  要: 目的研究维甲酸对支气管哮喘患者外周血辅助性T细胞17(Th17)/调节性T细胞(Treg)功能的调节作用。方法收集30例正常对照组,30例确诊支气管哮喘患者并在哮喘急性发作期的外周血标本,在无菌条件下,采用密度梯度离心法制备外周血单个核细胞(PBMC),将哮喘组PBMC置于含有不同浓度维甲酸(0.0、0.5、1.0、2.0μmo L/L)的培养基中,正常对照组PBMC置于不含维甲酸的培养基中,培养72 h后留取上清液,ELISA测定培养上清中IL-17、IL-10和转化生长因子-β(TGF-β)的水平。结果哮喘组PBMC培养基上清中的IL-17水平明显高于正常对照组(P<0.01),而IL-10和TGF-β的水平明显低于正常对照组(P<0.01);哮喘患者PBMC在维甲酸浓度为0.5、1.0、2.0μmo L/L培养基中培养后,上清中的IL-17水平均显著低于空白对照组水平(P<0.01),维甲酸的浓度越高,上清中的IL-17水平越低,但2.0μmo L/L组与1.0μmo L/L组之间的差异无统计学意义。哮喘患者PBMC在维甲酸浓度为0.5、1.0、2.0μmo L/L培养基中培养后,上清中的IL-10及TGF-β水平均显著高于空白对照组水平(P<0.01),维甲酸的浓度越高,上清中的IL-10及TGF-β水平也越高,但2.0μmo L/L组与1.0μmo L/L组之间的差异无统计学意义。结论维甲酸能抑制支气管哮喘患者外周血Th17功能,增强Treg细胞功能,为临床上治疗支气管哮喘提供新的方向及依据。 Objective To explore the regulation of retinoic acid on Th17/regulatory T cells (Treg) function in peripheral blood of patients with bronchial asthma, Methods Peripheral blood samples of 30 cases normal controls and 30 cases patients with bronchial asthma were col- lected. Under sterile conditions, peripheral blood mononuclear cells (PBMC) were prepared by density gradient centrifugation. PBMC were placed inculture medium of different concentra- tions of retinoic acid (0. 0, 0. 5, 1.0 and 2.0 μmoL/L). PBMC of normal control group were cultured without retinoic acid. Afterincubation of 72 h, the culture supematant were collected to determine IL-17, IL-10 and TGF- β level. Results IL-17 level in asthma group of PBMC medium was significantly higher than normal control group ( P 〈 0. 01 ), while the IL-10 and TGFβ levels were significantly lower than those of normal control group (P 〈 0.01 ) ; after in- cubation in culture medium of different concentrations of retinoic acid, IL-17 levels were signif- icantly lower than those of blank control group (P 〈0.01 ), but there was no significant differ- ence between 2.0 IxmoL,/L group and 1.0 IxmolML group. TGF-β levels were significantly higher than those of blank control group ( P 〈 0.01 ) , but there was no significant difference between 2. 0 ixmoL/L group and 1.0 ixmoL/L group. Conclusion Retinoic acid can inhibit Thl7 function in peripheral blood of patients with bronchial asthma, enhance Treg cell func- tion, and provide a new direction and basis for clinical treatment of bronchial asthma.
