摘 要: 目的:为了获得促进白木香(Aquilaria sinensis)形成沉香的真菌及其生物学特性。方法:采用PDA培养基分离并纯化真菌及菌液接种到白木香树进行功能验证。形态学特征结合ITS序列进行真菌鉴定和平板培养法测定真菌生物学特性。结果:分离到1株真菌ASAF01,其菌液输到白木香6个月后能促进沉香的形成。ASAF01鉴定为拟层孔菌(Fomitopsis sp.),属于拟层孔菌属(Fomitopsis),拟层孔菌科(Fomitopsidaceae),多孔菌目(Polyporales),伞菌纲(Agaricomycetes),担子菌门(Basidiomycota),其菌最适在马铃薯葡萄糖培养基上生长,最适温度是35℃,最适p H值为7,最适碳源和氮源为可溶性淀粉和酵母浸膏。结论:拟层孔菌(Fomitopsis sp.)ASAF01能促进白木香结香。 Objective:In order to screen a fungi to promote the agarwood formation in Aquilaria sinensis.Methods:The fungi were isolated by PDA medium,and then inoculated to A.sinensis to verify whether the fungi could promote agarwood formation.The fungi were identified based on morphological characteristics and DNA sequences analysis,then the biological characteristics observed by flat method.Results:ASAF01 strain could promote agarwood formation and was identified as Fomitopsis sp.,belonging to Fomitopsis,Fomitopsidaceae,Polyporale,Agaricomycetes,Basidiomycota.The results of biological characteristics showed that the mycelia of Fomitopsis sp.grew fastest in the PDA medium.The optimum temperature for the mycelial growth was 35 ℃ and the optimum pH value was 7.The optimal carbon sources and carbon source were soluble starch and yeast extract.Conclusion:Fomitopsis sp.ASAF01 can promote agarwood formation.
