摘 要: 目的:采用"双高优化法"优化柴胡多糖提取工艺,同时提高多糖得率和提取物多糖含量,降低部分难以经纯化除去的杂质。方法:在预实验筛选提取方法和醇沉工艺的基础上以水浴回流结合水提醇沉提取柴胡多糖,以"双高优化法"即以柴胡多糖得率和含量的归一值为评价指标,辅以响应面法优化出"双高"提取工艺,对"双高"工艺组和普通组分别作三次验证试验,测算两试验组的多糖含量差距,再对提取物进行初步纯化,研究两组多糖含量差异是否变化。结果:"双高"提取工艺:料也比38 m L·g^(-1),提取温度98℃,提取2次,每次4 h。验证结果:多糖得率6.23%,多糖含量53.57%,比普通优化组的含量高9.44%。用未筛选工艺的Sevag法初步纯化两组提取物,"双高"组精多糖含量达67.85%,与普通组精多糖含量差距不仅未缩小,反而有所增大。结论:经"双高优化法"优化的提取工艺,其多糖得率和提取物多糖含量均很高,该工艺能避免提取出部分无法由Sevag法除去的杂质,提高提取物在Sevag纯化中的纯化率。 Objective: "Combination optimization method" was applied to optimize Caihu polysaccharide extraction technology to improve the yield and content of polysaccharide with higher purity by reducing impurities which cannot be removed by purification. Method: On the basis of preliminary experimental screening extraction method and alcohol precipitation process, water bath reflux combined with water extract and alcohol precipitate were used to extract Caihu polysaccharide. Caihu polysaccharide yield and content were used as indexes and which combined the response surface method to optimize the extraction process. For validation, test were repeated three times and the polysaccharide content between two test groups were compared. Then a preliminary purification was conducted, and the content difference between the two groups were investigated. Result: The optimized extraction process by "Combination optimization method" were as following: ratio of material to solution 38 mL · g-1, temperature 98℃, extracting 2 times, each time 4 h. Caihu polysaccharide yield was 6.23% and content was 53.57% which was 9.44% higher than normal extraction process. Preliminary purification of the extracts of two groups by unscreened Scrag method, the refined polysaccharide content was 67.35% of the "Combination optimization method" group. The difference between the contents of refined polysaccharide content of the two groups increased. Conclusion: "Combination optimization method" in the optimization of the extraction process has high polysaccharide yield and content. The process can avoid extracting impurities which cannot by removed by Sevag purification method, and improve the purification rate of coarse extracts by Scrag method.
