摘 要: 目的:研究乙型肝炎病毒X蛋白(HBx)对肝癌细胞株中增殖分子、侵袭分子、血管新生分子表达的影响。方法:培养肝癌细胞株HepG2并分为HBx组和对照组,分别转染pcDNA3.1-HBx质粒以及空白的pcDNA3.1质粒。转染后24h和48h,检测细胞中增殖分子Survivin、cyclinD1、c-myc以及侵袭分子CD44v6、MT1-MMP、基质金属蛋白酶(MMP)2、MMP7以及血管新生分子血管内皮细胞生长因子(VEGF)、血管生成素(Ang)-1、Ang-2、成纤维细胞生长因子(FGF-2)的mRNA表达量。结果:转染24、48h后,HBx组细胞中Survivin、cyclinD1、c-myc、CD44v6、MT1-MMP、MMP2、MMP7、VEGF、Ang-1、Ang-2、FGF-2的mRNA表达量均显著高于对照组(P<0.05)。结论:HBx能够促进肝癌细胞株中增殖分子、侵袭分子、血管新生分子的表达。 Objective:To study the effect of hepatitis B virus X protein(HBx)on the expression of proliferation molecules,invasion molecules and angiogenesis molecules in liver cancer cell lines.Methods:Liver cancer cell lines HepG2 were cultured and divided into HBx group and control group.Liver cancer cell lines HepG2 were transfected with pcDNA3.1-HBx plasmid and blank pcDNA3.1plasmid respectively.A total of 24 hand 48hafter transfection,the mRNA expression of proliferation molecules Survivin,cyclinD1 and c-myc,invasion molecules CD44v6,MT1-MMP,MMP2 and MMP7as well as angiogenesis molecules VEGF,Ang-1,Ang-2and FGF-2in cells were determined.Results:After 24 hand 48hof transfection,the Survivin,cyclinD1,c-myc,CD44v6,MT1-MMP,MMP2,MMP7,VEGF,Ang-1,Ang-2and FGF-2 mRNA expression in HBx group of cells were significantly higher than those in control group.Conclusions:HBx can promote the expression of proliferation molecules,invasion molecules and angiogenesis molecules in the liver cancer cell lines.
