单纯室间隔缺损与房间隔缺损患者血清蛋白质组学分析

摘  要: 目的应用基质辅助激光解析/电离飞行时间质谱(MALDI-TOF-MS)技术对单纯性室间隔缺损(ventrieular septal defect,VSD)、房间隔缺损(atrial septal defect,ASD)患者术前、术后及正常人血清蛋白质谱进行分析,得到高敏感性和特异性的血清蛋白,建立诊断模型指纹图谱。方法采用MALDI-TOF-MS技术分别检测27例单纯VSD、ASD患者与20名正常人血清蛋白相对含量,运用Flex Analysis和Bio Tools软件对两组血清蛋白质谱进行分析;筛选出差异蛋白,再通过分析软件对其进行交叉验证,得到具有诊断意义的特异性蛋白标志物。结果检测到蛋白质质荷比(M/Z)在600~10500 Da范围内,单纯VSD、ASD组与正常人组有差异的蛋白质峰为15个(P<0.05)。生物标记物可以将单纯VSD、ASD组与正常人准确地分组,其敏感性为91.5%,特异性为97.2%。结论MALDI-TOF-MS技术对单纯VSD、ASD组术前、术后及正常人组血清蛋白质谱进行分析,可以筛选出单纯VSD、ASD患者的特异性血清标志物。 Obiective By using the technique of matrix assisted laser desorption/ionization-time of flight- mass spectrometry (MALDI-TOF-MS) to analyse the serum protein spectrum and to get high sensitivity and specificity protein marker in simple ventricular septal defect (VSD),atrial septal defect (ASD) patients (preoperative/after-operation) and normal persons,then to establish serum markers pattern for the diagnosis of VSD/ASD. Methods The technique of MALDI-TOF-MS was used to detect relative content of serum in 27 cases of VSD/ASD and 20 normal persons,and the Flex Analysis and BioTools software was used to analyse protein spectrum in two groups to find specific proteomic ,then through the analysis software for cross-validation to get a diagnosis from specific proteins. Resnlts There were 15 protein's peaks having significant discrepancy in the 600-10500Da (P〈0.05). The sensitivity and specificity was 91.5% and 97.2% respectively. Conclusion By using MALDI-TOF-MS to analyze serum protein spectrum in VSD/ASD group (preoperative/after-operation) and normal group could get the specific serum protein markers for diagnosis of VSD/ASD.
