建立快速光老化动物模型新方法
A new method for establishing animal model of rapid photoaging

摘  要: 目的寻找一种快速、有效的光老化动物模型制作途径。方法将20只雄性ICR小鼠随机分为正常对照组和模型组,每组各10只,使用UVA和UVB紫外线治疗仪对模型组进行照射,并逐渐增加剂量,最后第8周总计UVA和UVB照射总照射剂量为142.5 J/cm^2和20.52J/cm^2。照射结束后处死动物,对背部皮肤进行肉眼和微观形态学观察,并取组织匀浆行相关实验室检查,并进行统计学分析比较。结果模型组和正常对照组相比较肉眼外观及组织病理学、马松三色染色均出现了光老化表现;皮肤组织匀浆检测结果显示模型组与正常对照组相比GSH-px、SOD及HYP均明显降低,差异有统计学意义(P<0.05)。结论本次实验成功完成了快速动物光老化造模,这种方法值得推广。 Objective To find a rapid and effective method for making animal models of photoaging, develop a new approach quickly and effectively making photoaging animal model. Methods Twenty male ICR-mice were divided into two groups: 10 in normal control group and 10 in mould group. The model group was irradiated using UVA and UVB UV instruments and gradually increased doses, and total UVA and UVB exposure was 142.5 J/cm2 and 20.52J/ cm2 respectively at the last eighth weeks. The dorsal skin of two groups were observed. The tissue homogenate derived from the skin were investigaed and sta- tistically compared. Results Compared with the normal control group, the manifestation and histopathology of the model group and the trichrome staining of Masson showed photoaging in mould group; and the the laboratory rsuhs of GSH-px ,SOD and HYP in tissue homogenate of the mould group were lower sig- nificantly than the control group(P〈0.05 ). Conclusion This study has successfully and rapidly made photoaging animal model, which was worth promo- ting.
