摘 要: 目的通过测定结直肠癌患者癌组织、癌旁正常组织中mi R-196a和ZG16 m RNA的相对表达水平,探讨二者与患者临床病理特征的关系及二者相关性。方法应用实时荧光定量PCR,检测83例结直肠癌患者癌组织、癌旁正常组织中mi R-196a和ZG16 m RNA的相对表达水平,并分析二者相关性及其与患者临床病理特征之间的关系。结果癌组织中ZG16 m RNA表达量明显低于正常组织(t=-27.511,P<0.001);癌组织mi R-196a表达量明显高于正常组织(t=9.04,P<0.001),且二者在癌组织中的表达量呈显著负相关(P<0.001,r=-0.469);癌组织中mi R-196a、ZG16m RNA的表达情况与肿瘤部位、有无淋巴结转移、有无远处转移、肿瘤分期明显相关(P<0.05),与性别、年龄、肿瘤大小、分化程度、浸润深度、CEA水平无关(P>0.05)。结论两者可能推动结直肠癌的发生、发展,可能是潜在的病情评估及疗效评价的生物标志物;两者在左右半结直肠癌中表达量分别有差异,临床上可能有助于左右半结直肠癌靶向药物疗效预测及新的治疗靶点的发现。 Testing the expression of ZG16 m RNA and mi R-196 a of colorectal cancer tissue and the adjacent normal tissue to investigate the correlation between the two markers and detect their relationship with clinical pathological features of patients. Methods Quantitative real-time polymerase chain reaction(q RT-PCR) was used to detect the expression level of mi R-196 a and ZG16 m RNA in cancer tissue and its paired adjacent normal tissue sample from 83 CRC patients. The correlation of the two markers and their relative expression level with clinical-pathological features were also analyzed. Results The realative expression of ZG16 m RNA was significantly lower in CRC tissues compared with adjacent normal tissue samples(t=-27.511, P〈0.001); and the relative expression of mi R-196 a was significantly higher in cancer tissue compared with adjacent normal tissue samples(t=9.04, P〈0.001), and the relative expression of ZG16 m RNA has a significantly negative correlation with the relative expression of mi R-196a(P〈0.001, r=-0.469); The expression of mi R-196 a is related to tumor location, lymph node metastasis distant metastasis and tumor stages(P〈0.05), and its expression has no relationship with sex, age, the state of CEA, tumor size, differentiation degree or infiltration depthof CRC samples(P〉0.05). The same results was found in the expression of ZG16 m RNA. Conclusions Both of mi R-196 a and ZG16 m RNA may promote the occurrence of colorectal cancer, and suggests that they two may be potential biomarkers for disease and curative effect evaluation. Both of the expression of the two are different in left and right CRC, which may be helpful for individual treatment.
