microRNA-130a在慢性粒细胞白血病细胞耐药中的作用

摘  要: 目的比较microRNA-130a在伊马替尼耐药的慢性粒细胞白血病(CML)细胞株K562R和伊马替尼敏感的CML细胞株K562中的表达,探讨其与CML细胞耐药的关系。方法通过实时荧光定量PCR检测耐药株K562R和敏感株K562中microRNA-130a的表达水平。通过电穿孔转染法将microRNA-130a模拟物mimic转入敏感株K562中上调microRNA-130a的表达,分别用伊马替尼处理转染mimic细胞株及对照siRNA细胞株,记录各组细胞增殖情况。通过Annexin V和PI双染法分别检测各组细胞凋亡情况。使用不同浓度的伊马替尼处理转染mimic细胞株及对照细胞株,利用MTT法检测各组细胞对伊马替尼的敏感性。结果 microRNA-130a在耐药株K562中的表达明显高于敏感株K562。转染microRNA-130a模拟物mimic的K562细胞对伊马替尼的增殖抑制率明显低于对照组,凋亡情况也明显少于对照组。使用不同浓度的伊马替尼处理转染mimic细胞株及对照细胞株,mimic组细胞增殖抑制率明显低于对照组,IC_(50)分别为0.13μmol/L和3.19μmol/L。结论 microRNA-130a在伊马替尼耐药株K562 R中呈高表达,上调microRNA-130a的表达可降低K562对伊马替尼的敏感性。 Objective To compare the expression of microRNA-130a in imatinib-resistant K562R cells and imatinib-sensitive K562 cells,and to investigate the relationship between microRNA-130a and drug resistance of chronic myeloid leukemia(CML).Methods The expression of microRNA-130a in imatinib-resistant K562 R cells and imatinib-sensitive K562 cells was detected by real-time RTPCR.M icroRNA-130a mimic was transferred into K562 cells by electro-transfection.K562/mimic and the control cells were treated with imatinib,then the cell proliferation was recorded and apoptosis was detected by flow cytometry with Annexin V/PI staining.M TT method was used to detect the proliferation ability of K562/mimic and the control cells treated with imatinib.Results The expression level of microRNA-130a in imatinib-resistant K562 R cells was higher than that of imatinib-sensitive K562 cells.The proliferation inhibition rate to imatinib and the apoptosis rate of K562/mimic were lower than those of control cells,and the IC50 was 0.13 μmol/L and 3.19 μmol/L respectively.Conclusion MicroRNA-130a is highly expressed in imatinib-resistant K562 R cells,and up-regulating the expression of microRNA-130a can decrease the sensitivity of imatinib to CML.
