摘 要: 目的检测G蛋白αq亚单位(Gαq)在寻常型银屑病患者外周血单个核细胞(PBMC)中的mRNA表达水平,阐明Gαq与寻常型银屑病发病或疾病活动程度的相关性。方法搜集45例寻常型银屑病患者外周血10mL,采用银屑病面积和严重度指数(PASI)评分法进行病情严重程度评分,以46例匹配的正常人为对照。荧光定量PCR检测PBMC中Gαq蛋白的表达水平。同时检测血清炎症指标CRP。结果与健康对照相比,Gαq的mRNA水平在寻常型银屑病患者的外周血单个核细胞中的表达明显降低(P<0.05),与中重度银屑病的活动度(PASI评分、C反应蛋白水平)有显著相关性(r分别等于-0.515和-0.458)。结论 Gαq可能作为负调节因素参与银屑病的发病。 Objective To detect the expression level of Gαq mRNAin peripheral blood mononuclear cell(PBMC)of patients with psoriasis vulgaris,and to clarify the relationship between Gαq and disease or disease activity of psoriasis vulgaris.Methods Thirty cases with psoriasis vulgaris were collected,after informed consent,the peripheral blood 10 mL were taken,the disease severity score were peformed according to the PASI scores method,30 cases of the matched normal were collected as controls.PBMC were separated by Ficoll-hypaque density gradient centrifugation.Fluorescence quantitative PCR detected Gαq protein expression level.To test some inflammatory indexes which associated with disease activity or development,and then analysed the corelation between Gαq expression and disease activity,serological indicator of inflammation.Results Our data showed that the expressions of G alpha q protein and mRNA in peripheral blood mononuclear cells of patients with psoriasis vulgaris were significantly lower than those of healthy controls.There was a negative correlation between the expression of G alpha q and disease activity in moderate and severe psoriasis vulgaris.Conclusions Galphaq probably plays a vital role in the pathogenesis of psoriasis vulgaris as a negative regulator.
