骨肉瘤组织中Runt相关转录因子3表达量与细胞增殖、血管新生的相关性

摘  要: 目的:探讨骨肉瘤组织中Runt相关转录因子3(RUNX3)表达量与细胞增殖、血管新生的相关性。方法:收集2014年2月~2017年2月间在本院接受治疗的骨肉瘤患者80例,检测肿瘤组织及肉瘤旁组织中RUNX3的表达量。进一步根据肿瘤组织RUNX3表达量分为RUNX3高表达组、RUNX3低表达组,对比其增殖基因、血管新生基因表达量的差异。结果:骨肉瘤组织中RUNX3、KISS-1、RanBP9mRNA表达量显著低于肉瘤旁组织,VCP、Six1、S100A6、IF-1α、MMP-14、bFGF、Ang-2mRNA表达量显著高于肉瘤旁组织;RUNX3高表达组骨肉瘤组织中KISS-1、RanBP9 mRNA的表达量高于RUNX3低表达组,VCP、Six1、S100A6、HIF-1α、MMP-14、bFGF、Ang-2mRNA mRNA的表达量低于RUNX3低表达组。结论:骨肉瘤组织中RUNX3表达量减少,是导致肿瘤增殖活性增高、血管新生旺盛的直接原因之一。 Objective:To study the correlation of Runt-related transcription factor gene 3(RUNX3)expression in osteosarcoma tissue with cell proliferation and angiogenesis.Methods:A total of 80 patients with osteosarcoma who were treated in our hospital between February 2014 and February 2017 were collected,and RUNX3 expression in osteosarcoma tissue and adjacent tissue were detected.According to RUNX3 expression in tumor tissue,the 80 patients were further divided into high RUNX3 expression group and low RUNX3 expression group,and proliferation gene and angiogenesis gene expression of two groups were compared.Results:RUNX3,KISS-1and RanBP9 mRNA expression in osteosarcoma tissue were significantly lower than those in adjacent tissue(P〈0.05)while VCP,Six1,S100A6,IF-1α,MMP-14,bFGF and Ang-2mRNA expression were significantly higher than those in adjacent tissue(P〈0.05);KISS-1and RanBP9 mRNA expression in osteosarcoma tissue of high RUNX3 expression group were significantly higher than those of low RUNX3 expression group(P〈0.05)while VCP,Six1,S100A6,IF-1α,MMP-14,bFGF and Ang-2mRNA expression were significantly lower than those of low RUNX3 expression group(P〈0.05).Conclusions:The decrease of RUNX3 expression in osteosarcoma tissue is one of the direct causes of the increased tumor proliferation activity and strong angiogenesis.
