复方蛹虫草对小肠炎症和IEC-6存活率的干预作用
Effects of compound Cordyceps militaris on small intestine inflammation and the survival rate of IEC-6 cells

摘  要: 目的探究复方蛹虫草(CCM)对高脂诱导的小肠炎症和H_2O_2与LPS诱导的IEC-6细胞凋亡的干预作用。方法C57BL/6J雄性小鼠随机分为正常对照组、模型组及CCM 90、180和540 mg·kg^(-1)·d^(-1)组。采用高脂饮食建立小鼠小肠炎症模型。造模同时ig给药,每天1次,连续8周,比色法测定TG、T-CHO、LDL-C、HDL-C;酶联免疫吸附测定法(ELISA)测定血清细胞炎性因子TNF-α、IL-1β、IL-6、LPS。HE染色法比较各组空肠、回肠组织病理改变,MTT方法检测H_2O_2与LPS诱导的IEC-6细胞存活率。结果与模型组相比,CCM各给药组T-CHO、LDL-C水平显著降低;小肠肠腺数目显著增加,黏膜层厚度接近正常组小肠组织,黏膜上皮细胞未见明显变性、坏死,炎性细胞消失;CCM给药组小鼠血清TNF-α、IL-1β、IL-6、LPS含量显著降低;CCM各组IEC-6细胞存活率显著增加且效果好于CME组和GBE组。结论 CCM可以改善高脂诱导的小鼠小肠炎症,其机理可能与其提高小肠上皮细胞的存活率有关。 This study was performed to explore the intervention effect of compound Cordyceps militaris(CCM)on small intestinal inflammation induced by high-fat and on IEC-6 cell apoptosis induced by LPS and H2O2. The C57BL/6J mouse model of small intestinal inflammation was induced with high-fat diet, followed by oral administration of 90, 180 or 540 mg·kg(-1)·d(-1)CCM, once a day for 8 weeks. Then we observed the levels of total cholesterol(TG), triacylglyceride(T-CHO), low density lipoprotein cholesterol(LDL-C), high density lipoprotein cholesterol(HDL-C), tumor necrosis factor-α(TNF-α), interleukin-1β(IL-1β), interleukin 6(IL-6),lipopolysaccharide(LPS) in serum, as well as pathological changes in jejunum and ileum tissue. The survival rate of IEC-6 cells induced by H2O2 and LPS was determined by MTT method. Compared with the model group, CCM groups showed obvious relieves in inflammatory infiltration in mouse jejunum and ileum, significant reduction in T-CHO, LDL-C, TNF-α, IL-1β, IL-6 and LPS in serum, and CCM can increased cell survival rate significantly. In conclusion, CCM can improve mouse small intestine inflammation induced by high fat, and the mechanism may associate with the inhibition of small intestine epithelial cells apoptosis.
