葛根素对STZ糖尿病模型大鼠视网膜VEGF的调节及对视网膜细胞凋亡的促进作用的研究

摘  要: 目的研究葛根素对链脲佐菌素(STZ)糖尿病大鼠视网膜血管内皮生长因子(VEGF)的调节及对视网膜细胞凋亡的促进作用。方法清洁级SD大鼠随机分为实验组及对照组各28只,实验组一次性腹腔注射1%STZ诱发糖尿病模型,对照组腹腔注射磷酸盐缓冲液(PBS)。成模后给两组大鼠分别注射葛根素,4、8、16、24周后采用实时荧光定量(Real-time PCR)观察VEGF的表达水平,流式细胞技术检测细胞凋亡,WB检测凋亡相关蛋白磷酸化AKT(pAKT)及Caspase3、8、9表达量。结果与对照组相比,实验组大鼠注射葛根素4、8周后VEGF表达量,视网膜组织细胞凋亡,pAKT、Caspase8、Caspase9及Caspase3表达均改变不明显,差异无统计学意义(P>0.05);但16、24周后VEGF表达量发生明显下降(P<0.05),视网膜组织细胞凋亡发生明显升高(P<0.05),pAKT表达下调,Caspase8、Caspase9及Caspase3表达均上调(P<0.05)。结论较长时间注射葛根素能下调糖尿病大鼠视网膜VEGF的表达,从而下调pAKT表达,最终导致Caspase3、8、9的表达上调,诱导细胞发生凋亡。此研究结果为糖尿病性视网膜病变的治疗提供了实验依据。 Objective To explore the regulation effect of regulation on STZ diabetic rats retinal VEGF and the promoting apoptosis study of puerarin on STZ diabetic rats retinal. Methods Sprague - Dawley (SD) rats were randomly divided into experimental group and control group, 28 rats in each group. The experimental group receiving 1% intraperitoneal injection of STZ induced diabetic model, the control group re- ceiving intraperitoneal injection of PBS. The successful model were injected puerarin 4, 8, 16, 24 weeks . And real - time PCR was used to ob- serve the expression of VEGF level, flow eytometry technology was used to detect cell apoptosis, WB to detect apoptosis related proteins phospho- rylated AKT and Caspase3, 8, 9. Results Compared with control group, experimental group rats after puerarin injection, the VEGF expression of change at 4, 8 weeks is not obvious, apoptosis , and phosphorylated AKT and Caspase 3,8, 9 are not obvious, there were also no significant change. 16 weeks after injection of puerarin, the amount of VEGF expression bad obvious changes and apoptosis is also more obvious, the expres- sion of phosphorylated AKT and Caspase 3 and 8, 9 also has the obvious changes, but 24 weeks after injection of puerarin the level of VEGF ex- pression and apoptosis is also very obvious, phosphorylated AKT and the expression of Caspase 3,8,9 had obvious change. Conclusion Puerar- in injection for a long time can decrease the expression of vascular endothelial growth factor VEGF, so as to decrease the phosphorylated AKT, e- ventually result in the expression of Caspase3, 8, 9 up - regulation, inducing cell apoptosis occuring. The research results provides the experimen- tal basis for treatment of STZ diabetic rats retina.
