摘 要: 目的观察大蒜素对慢性肾衰(CRF)大鼠贫血和血脂、电解质代谢紊乱的影响。方法 100只实验用大鼠随机分为正常对照组、模型对照组、大蒜素[5、10、20 mg/(kg·d)]组,采用腺嘌呤溶液[250 mg/(kg·d)]灌胃21 d的方法建立CRF大鼠模型,大蒜素[5、10、20 mg/(kg·d)]组腹腔注射给药治疗,疗程28 d。测定血清肾功能指标(BUN、SCr、UA含量),计算肾脏指数;测定全血中红细胞(RBC)、血红蛋白(HGB)、血球压积(HCT)、血清铁(SI)、总铁结合力(TIBC)、铁蛋白(FER);测定血浆中甘油三酯(TG)、总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)含量;测定血清中钾(K^+)、钠(Na^+)、钙(Ca^(2+))、磷(P^(3+))含量;HE染色观察肾脏组织病理变化。结果与模型对照组比较,大蒜素10、20 mg/(kg·d)组CRF大鼠血清BUN、SCr、UA含量显著降低(P<0.05或P<0.01),肾脏指数显著降低(P<0.01),HGB、HCT及SI、TIBC、FER均显著升高且20 mg/(kg·d)组RBC显著升高(P<0.05或P<0.01),血浆中TG、TC、LDL-C含量水平显著降低且20 mg/(kg·d)组HDL-C含量水平显著升高(P<0.05或P<0.01),血清中Ca^(2+)含量显著升高(P<0.05),P^(3+)含量显著降低且20 mg/(kg·d)组血清K^+、Na^+显著降低(P<0.05或P<0.01)。结论大蒜素可能通过改善贫血、电解质紊乱及调节血脂而对CRF大鼠起到一定的保护作用。 Objective: To study the effect of allicin on anemia,blood fat and electrolyte metabolism in rats with chronic renal failure(CRF). Methods: 100 rats were randomly divided into the normal control group,the model control group and allicin[5,10,20 mg/(kg·d)] groups. CRF rat model was established by intragastric administration of adenine solution [250 mg/(kg·d)] for 21 days,and then intraperitoneal injection of allicin [5,10,20 mg/(kg·d)] for treatment,once a day for 28 days. The levels of serum urea nitrogen(BUN),serum creatinine(SCr)and uric acid(UA) were measured;the kidney index was calculated. The level of red blood cells(RBC),hemoglobin(HGB),hematocrit(HCT),serum iron(SI),total iron binding capacity(TIBC) and ferritin(FER) in whole blood were measured;the content of triglyceride(TG),total cholesterol(TC),low density lipoprotein cholesterol(LDL-C) and high density lipoprotein cholesterol(HDL-C) in plasma were measured;the level of potassium(K~+),sodium(Na~+),calcium(Ca^(2+)) and phosphorus(P^(3+)) in serum were measured. The pathological changes of renal tissue was observed by HE staining. Results: Compared with the model group,the content of BUN,SCr,UA in serum of allicin[10,20 mg/(kg·d)] groups were significantly reduced(P 0.05 or P 0.01);renal index was significantly reduced(P 0.01);the level of HGB,HCT,SI,TIBC and FER was significantly increased and RBC of 20 mg/kgod group increased significantly(P 0.05 or P 0.01);the content of TG,TC and LDL-C in plasma was significantly decreased and the HDL-C in the 20 mg/(kg·d) group was significantly increased(P 0.05 or P 0.01);the level of Ca^(2+) in serum was significantly increased(P 0.05) and the level of K~+,Na~+ and P^(3+) were significantly decreased(P 0.05 or P 0.01). Conclusion: Allicin perhaps has protective effects on CRF rats by improving anemia and electrolyte imbalance and regulating blood lipids.
