摘 要: 目的观察姜黄素对蛛网膜下腔出血大鼠脑血管功能的影响及血清血小板源性生长因子(PDGF)、血管内皮生长因子(VEGF)、表皮生长因子受体(EGFR)的变化。方法将60只雄性SD大鼠随机分为假手术组、模型组、姜黄素治疗组(姜黄素组),各20只。通过枕大池二次注血法建立蛛网膜下腔出血模型。所有造模成功的大鼠于第1次注血30 min后开始给药,具体方法如下:假手术组、模型组给予生理盐水;姜黄素组给予姜黄素2 m L。3组大鼠均采用灌胃的方式,连续给药7 d。术前、给药后1、3、5、7 d使用神经学评分评估大鼠神经功能。于给药后7 d取基底动脉进行HE染色,观察基底动脉横截面积和血管壁厚度。放射性免疫法分别测各组大鼠血清PDGF、VEGF、EGFR水平。结果与假手术组相比,模型组神经学评分在给药1 d、3 d、5 d、7 d后均显著增加,差异有统计学意义(P<0.05),而姜黄素组评分虽高于假手术组,但差异无统计学意义(P>0.05)。与模型组相比,姜黄素组评分在给药1 d、3 d、5 d、7 d后均显著下降,差异有统计学意义(P<0.05)。基底动脉HE染色发现,模型组基底动脉发生管腔狭窄,壁厚增加,内皮细胞扭曲,内部弹性层卷积等形态学变化,而姜黄素组皱缩及增厚程度减轻。与假手术组相比,模型组横截面积均显著降低,血管壁厚度显著增加(P<0.05)。而与模型组相比,姜黄素组横截面积显著增加,血管壁厚度显著降低(P<0.05)。而假手术组与姜黄素组差异均无统计学意义(P>0.05)。放射性免疫法检测结果显示,与假手术组相比,模型组PDGF、VEGF、EGFR水平显著增加(P<0.05)。而与模型组相比,姜黄素组PDGF、VEGF、EGFR水平显著下降(P<0.05)。结论姜黄素能改善蛛网膜下腔出血大鼠脑血管功能,并降低血清PDGF、VEGF、EGFR的表达。 Objective: To investigate the effect of curcumin on cerebral vascular function and serum PDGF,VEGF and EGFR in rats with subarachnoid hemorrhage. Methods: 60 male SD rats were randomly divided into three groups: the shame group,the model group,and curcumin group(n = 20 in each group). A SAH model was induced by injection of blood twice into cisterna magna. At 30 min before and after the two blood injictions,the shame group and the model group were given saline;curcumin group were given curcumin 2 m L(curcumin with distilled water prepared into 10 g/kg of the liquid). Three groups of rats were fed with 7 days of continuous administration. Neurological score was used to assess neurological function in rats when preoperative and postopera-tive 1,3,5,7 days. HE staining was used to observe the diameter and cross-section area at day7 after dosing. The levels of serum PDGF,VEGF and EGFR were measured by radioimmunoassay. Results: Compared with the shame group,the neurological score of the model group was significantly increased after 1 d,3 d,5 d and 7 d.Compared with The model group,the score of curcumin group in the administration 1 d,3 d,5 d,7 d after the significant decreased(P 0.05). The morphological changes of the model group were observed including luminal narrowing,increased wall thickness,distorted endothelial cells,and convolution of the internal elastic lamina,while slight decreased vasospasm was seen in the curcumin group. Histograms of the average cross-sectional area of basilar artety showed significant decrease and the average wall thickness increased in the model group,compared with that in the shame group,which was attenuated by curcumin group. The levels of PDGF,VEGF and EGFR in the model group were significantly increased. Compared with the model group,the levels of PDGF,VEGF and EGFR in curcumin group were significantly decreased. Conclusion: Curcumin can improve the cerebrovascular function of rats with subarachnoid hemorrhage,and decrease the expression of serum PDGF,VEGF and
