TGF-β1 Smad7和Ki67在星形细胞瘤中的表达及其相关性

摘  要: 目的:观察正常脑组织以及不同分级脑星形细胞瘤中TGF-β1、Smad7和Ki67的表达。方法:经病理确诊的星形胶质细胞瘤患者50例,其中Ⅰ-Ⅱ级患者20例(Ⅰ-Ⅱ级组),Ⅲ-Ⅳ级患者30例(Ⅲ-Ⅳ级组),取手术切除的肿瘤组织及以同期脑外伤行减压术切除的额颞叶正常脑组织(对照组,10例),采用免疫组织化学法和RT-PCR分别检测标本脑组织切除的中TGF-β1、Smad7和Ki67蛋白及mRNA表达,并分析星形胶质细胞瘤中3项指标的相关性。结果:在星形胶质细胞瘤中,TGF-β1、Smad7和Ki67蛋白及mRNA表达水平明显高于对照组(P<0.01),且随着星形细胞瘤临床病理分级的升高而升高(P<0.01);在星形胶质细胞瘤中,TGF-β1与Smad7呈正相关(r=0.76,P<0.01),TGF-β1与Ki67呈正相关(r=0.74、0.72,P<0.01),Smad7与Ki67呈正相关(r=0.72,P<0.01)。结论:TGF-β1、Smad7和Ki67在星形胶质细胞瘤中表达水平明显高于正常脑组织,且随肿瘤病理分级增高而增加,3项指标互成正相关关系。 Objective: To explore the normal brain tissue and the expression of TGF-β1, Smad7 and Ki67 proteins in the different grade astroeytoma. Methods :There were 50 patients with pathologically confirmed astrocytomas, of whom 20 patients were grade Ⅰ-Ⅱ ( Group Ⅰ-Ⅱ), and 30 patients were grade Ⅲ-Ⅳ (Group Ⅲ-Ⅳ ). The tumor tissue and frontotemporal normal brain tissue reseeted during trauma decompression surgery in the same period (control group, 10 cases) were collected. Immuno- histochemistry and RT-PCR were used to detect TGF-β1, Smad7, Ki67 protein and mRNA expression in the brain tissues to analyze the correlation of 3 indexes in astrocytomas. Results: In astrocytomas, the expression levels of TGF-β1, Smad7, Ki67 proteins and mRNA were significantly higher than those of control group, and these indexes increased with the increase of clinical pathological grading of astro- cytoma (P 〈 0. 01 ). In astrocytomas, TGF- beta 1 is positively related to Smad7 ( r = 0.76, P 〈 0. 01 ) ; TGF-β1 is positively related to Ki67 ( r = 0.74, P 〈 0.01 ) ; Smad7 is positively related to Ki67 ( r = 0.72 ,P 〈 0. 01 ). Conclusion: The expression levels of TGF-β1, Smad7 and Ki67 in astro cytomas were significantly higher than those in normal brain tissues, and these indexes increased with the increase of tumor grade, and the 3 indexes are positively correlated with each other.
