甲状腺功能减退对雄性大鼠睾丸线粒体ERα和AR基因mRNA表达的影响

摘  要: 目的探讨甲状腺功能减退(甲减)对雄性大鼠睾丸线粒体性激素受体(ERα和AR)基因mRNA表达的影响,为研究甲减对雄性生殖系统损伤机制提供理论依据。方法 30只健康雄性wistar大鼠,随机分3组,按体质量,采用随机数字表法分为3组:对照组(C组,灌胃生理盐水1 ml/100 g)、低剂量组(L组,灌胃0.1 mg/kg丙硫氧嘧啶1 ml/100 g)和高剂量组(H组,灌胃10 mg/kg丙硫氧嘧啶1 ml/100 g),每组10只,灌胃60 d,每隔2天称体重1次。灌胃结束次日处死大鼠,放射免疫法检测血清中甲状腺激素(TT3、TT4和TSH);实时荧光定量PCR(RT-PCR)法检测睾丸线粒体ERα和AR基因mRNA相对表达量。结果与C组和L组比较,H组大鼠体重、TT3和TT4出现明显降低(F=19.82,P<0.05;F=139.34,P<0.05),TSH水平及睾丸脏器系数显著升高(F=60.389,P<0.05;F=21.83,P<0.05);基因结果显示:H组ERα基因mRNA相对表达量显著高于C组和L组(F=13.35,P<0.05),AR基因mRNA相对表达量显著性高于C组(F=4.561,P<0.05)上述差异均有统计学意义。结论甲减可影响性激素受体基因mRNA表达,进而损伤大鼠生殖系统。 Objective To define the effect of hypothyroidism on the mRNA expressions of sex hormone receptors( ERα and AR) in testicular mitochondria of male rats to provide a theoretical basis for the mechanism of male reproductive system damage. Methods According to body weight,30 wistar male rats were divided into control group( C group,1 ml/kg·d NS by intragastric administration),Lowdose group( L group,0. 1 mg/kg propylthiouracil1 1 ml/kg·d by intragastric administration) and high-dose group( H group,10 mg/kg propylthiouracil1 1 ml/kg·d by intragastric administration). After 60 days,all rats were sacrificed. The levels of thyroid hormones( TT3,TT4 and TSH) were measured by radioimmunoassay. The mRNA expression levels of ERα and AR in testicular mitochondria were detected by real-time quantitative PCR( RT-PCR). Result Compared with C and L group,the levels of TT3,TT4 and weights were significantly decreased in H group( F = 19. 82,P〈0. 05; F = 139. 34,P〈0. 05),whereas the level of TSH and testicular organ coefficient were increased in H group( F = 60. 389,P〈0. 05; F = 21. 83,P〈0. 05). The result of RT-PCR showed that the relative mRNA expression of Erα was significant increased compared with C and L groups( F = 13. 35,P〈0. 05) and the relative mRNA expression of AR was significant increased compared with C group( F = 4. 561,P〈0. 05) in H group. Conclusion Hypothyroidism may induce the change of mRNA expression of sex hormone receptors and then damage the reproductive system in male rats.
