不同培养条件对黄瓜发状根生长影响研究

摘  要: 本研究以PCR分子鉴定转化成功的黄瓜发状根为试材,采用悬浮培养的方法分析不同培养基类型、不同培养基组分和不同激素对黄瓜发状根生长的影响。分析显示:MS培养基中黄瓜发状根生长速度最快,其次为1/2 MS培养基,N6培养基中发状根生长的速度最慢;以MS为基础培养基,30 g/L的蔗糖为氮源,C_(NH4+):C_(NO3)-=1:2为碳源和204 mg/L KH_2PO_4为磷源时,培养基中黄瓜发状根生长量最大;以MS为基础培养基,分别添加激素IAA(Indole acetic acid)、GA3(Gibberellin)、NAA(Naphthalene acetic acid)、KT(Kinetin),其中1 mg/L IAA作用下的黄瓜发状根的增长量最大(51.0 mg),1 mg/L的NAA(Naphthalene acetic acid)处理下黄瓜发状根的增长量最少(37.8 mg/L)。以上为发状根进一步扩大培养和后期的工业化大规模生产提供一定理论依据。 Hairy roots of cucumber with identified by PCR were used as experiment material in this study. The effects of different media types, different media components and different hormones on the growth of hairy roots in cucumber were analyzed by methods of suspension culture. The analysis showed that the growth rate of cucumber hairy root was the fastest in MS medium, followed by 1/2 MS medium and the growth rate of hairy root was the slowest in N6 medium. MS medium has been applied to the basal medium, the 30 g/L sucrose as the sole carbon source, C_(NH4+):C_(NO3)-=1:2 as the carbon source and the 204 mg/L KH_2PO_4 solution as the phosphorus source, the growth of cucumber hairy root was the highest in the medium. The hormones IAA(Indole acetic acid), GA3(Gibberellin), NAA(Naphthalene acetic acid) and KT(Kinetin) were added, respectively. The growth of cucumber hairy roots with the 1 mg/L IAA was the largest(51.0 mg), and with the 1 mg/L NAA was the least(37.8 mg/L).These results provide a theoretical basis for the further expansion of hairy roots and the later industrialization of large-scale production.
