马铃薯品种Favorita再生体系的优化
Optimization of Regeneration System from Potato Variety Favorita

摘  要: 以马铃薯品种Favorita脱毒试管苗的叶片与茎段为外植体,在愈伤诱导培养基上培养不同时间,然后转入到添加了不同浓度6-BA与ZTR组合的芽诱导培养基上,观察其对马铃薯愈伤组织诱导及芽分化的影响。结果表明:茎段与叶片在愈伤培养基(MS+1 mg/L IAA+0.2 mg/L GA3+0.5 mg/L 6-BA)上分别培养2 d、4 d、6 d后,再转入添加不同浓度6-BA和ZTR的芽诱培养基上,均可以100%产生愈伤组织;50 d后调查,诱导2 d的叶片组织转入添加1 mg/L IAA、0.2 mg/L GA3、2 mg/L 6-BA和1 mg/L ZTR的MS诱导培养基,再生率100%,诱导6 d的叶片组织转入添加1 mg/L IAA、0.2 mg/L GA3、0.5 mg/L 6-BA和2 mg/L ZTR,分化率100%;诱导2 d的茎段组织转入添加1 mg/L IAA、0.2 mg/L GA3、3 mg/L 6-BA和1 mg/L ZTR,分化率50%。 Using potato variety Favorita virus-free plantlets' leaves and stem segments as explant, which are cultivated in callus induction medium in different timings, then the virus-free plantlets' leaves and stem segments are transplanted to bud induction medium having added different concentration of 6-BA combined with ZTR, to observe the effect for the callus induction and bud differentiation of potato. The results show that: Segments of stem and leaves which are cultivated on callus induction medium(MS +1 mg/L IAA+0.2 mg/L GA3+0.5 mg/L6-BA) for two days, four days, six days, respectively, and then transferred to shoot-inducing medium with different concentrations of 6-BA and ZTR, can form callus for 100%. After 50 days, the investigation shows that the regeneration rate is 100% for the leaves which are cultivated on callus induction medium two days, and then transferred to shoot-induction medium with 1 mg/L IAA, 0.2 mg/L GA3, 2 mg/L 6-BA and 1 mg/L ZTR. The differentiation rate is 100% for the leaves which are cultivated on callus induction medium for six days, and then transferred to shoot-induction medium with 1 mg/L IAA, 0.2 mg/L GA3, 0.5 mg/L 6-BA and 2 mg/L ZTR. The differentiation rate is 50% for the leaves which are cultivated on callus induction medium for two days, and then transferred to shoot-induction medium with 1 mg/L IAA, 0.2 mg/L GA3, 3 mg/L 6-BA and 1 mg/L ZTR.
