一株松鼠猴源粪肠球菌的分离鉴定及耐药性分析

摘  要: 为了确定引起松鼠猴死亡的致病菌株及其耐药性情况,试验采用病原菌分离、革兰氏染色、生化试验、16S rRNA PCR扩增鉴定、致病性试验、药敏试验、毒力基因和耐药基因扩增试验等方法对致病菌进行了研究。结果表明:该菌革兰氏染色阳性,马尿酸盐、V-P、6.5%Na Cl等生化反应呈阳性,阿拉伯糖、蔗糖等生化反应呈阴性;16S rRNA基因BLAST比对,与粪肠球菌相似性为100%;致病性试验显示,该菌株致病性不是很强;该菌株对苯唑西林、红霉素、复方新诺明、四环素等耐药,对青霉素G、呋喃妥因敏感,对万古霉素、米诺环素中等耐药;毒力基因hyl、asal、gelE、cylA、esp、acm和耐药基因TEM、ant(6)-I、ermB、aac(6')-aph(2″)、TetM扩增试验均为阳性。说明该菌株为粪肠球菌(Enterococcus faecalis),具有较强耐药性,但致病性不是很强,可能与毒力基因和耐药基因的高阳性率有关。 To identify the pathogenic Strains which led to the death of Saimiri sciureus and their antibiotic resistance, a variety of methods for bacteria detection were used in this experiment includingpath^gen isolation, Gram staining, biochemical test and 16S rRNA based PCR amplification, animalpathogenicity test, drug susceptibility testand PCR amplificationfor virulence genes and resistance genes. The results showed that the strain was Gram- positive as well as positive inreaetionwith hippurate, V- P,6.5 % NaCl and etc;it was negative in reactionwitharabinose, su- croseand etc. The strain showed 100% similarity with Enterococcusfaecalis. Animalpathogenicity test showed that the strain was not highlypatho7 genic. The strain was highly resistant to oxacillin, erythromycin, compound sinomin, tetracycline, while was sensitive to penicillin G and nitrofurantoin, and moderately resistant to vancomycin, minocyeline. PCR amplification results showed positive in the strain including virulence gene shyl, asal, gelE, cylA, esp, acm, and resistance genes TEM, ant ( 6 ) - I, ermB, aac (6′) - aph ( 2″), TetM. The results indicated that the bacteria was Enterococcus faecalis, with high drug resistance and low pathogenicity, which could be in relation to highly positive rates of virulence genes and resistance genes.
