摘 要: 建立HPLC法用于测定核苷酸的含量。亲水柱HILCI为色谱柱,以磷酸二氢钾(0.01 mol/L)-乙腈(22︰78,V/V)为流动相,检测波长为260 nm;在这一色谱条件下,五种核苷酸分离良好。测得五核苷酸标准曲线的相关系数均大于0.999 4,回收率在95.3%~104.2%之间。该法简单、灵敏、准确、精密满足检测需求。 A HPLC method for the determination of nucleotide in milk products for infants and young children had been established. The samples were analyzed by the separation on a HILIC chromatographic column with an 0.01 mol/L potassium phosphate monobasic-acetonitrile (22 : 78, V/V) as the mobile phase and the determination with ultraviolet detector at 260 nm. The good correlation coefficients of the analytes were obtained within their respective linear ranges. A linearity with a good correlation coefficient was r=0.999 4, the average recovery was 95.3%- 114.2% This method was simple, sensitive, accurate and precise, and could be used for determination of Nucleotide in milk products for infants and young children.
