富氢水对4 Gy X射线全身照射小鼠造血功能的影响

摘  要: 目的观察富氢水对4 Gy全身照射的小鼠造血功能的影响。方法将30只C57BL/6小鼠分为对照组、照射组及富氢水组,对照组接受假照射,照射组小鼠给予4 Gy X射线全身照射,富氢水组小鼠4 Gy X射线全身照射前2 h及照射后每天给予20 ml/kg富氢水灌胃。照射前和照射后1、4、7、10、14 d割尾取血20μl,通过血细胞分析仪检测外周血红细胞、白细胞、中性粒细胞及血小板计数。照射后第6天,无菌条件下取小鼠单侧股骨骨髓细胞接种,计数粒细胞-巨噬细胞系集落形成单位(CFU-GM)。采用独立样本t检验检测每组细胞数在不同时间点的变化。结果照射后照射组及富氢水组外周血红细胞在第14天达到最低值,分别为(5.0±0.1)31012/L和(7.3±0.2)31012/L;照射组及富氢水组外周血白细胞第4天达到最低值,分别为(4.7±0.7)3109/L和(9.4±1.2)3109/L;两组外周血中性粒细胞第4天达到最低值,分别为(1.4±0.4)3109/L和(3.0±0.1)3109/L;两组外周血血小板第7天达到最低值,分别为(200.0±27.0)3109/L和(300.0±41.0)3109/L。照射组与富氢水组比较,富氢水组小鼠全身照射后外周血细胞下降程度更小,恢复时间更短。第6天,富氢水组CFU-GM(124.0±8.4)3105/L,而照射组为(64.0±7.4)3105/L,两者间有统计学差异(P<0.05)。结论富氢水可以减轻4 Gy X射线全身照射的小鼠的骨髓抑制。 Objective To observe the effect of hydrogen rich water on hematopoietic function in mice exposed to 4 Gy whole body irradiation. Methods 30 C57BL/6 mice were divided into control group, radiation group and hydrogen rich water group, the control group received sham irradiation, irradiated group mice were given 4 Gy X ray irradiation, hydrogen rich water group 4 Gy X radiation 2 h before and after irradiation(20 ml/kg) given daily intragastric administration of hydrogen rich water. Before irradiation and after irradiation for 1 days, 4 days, 7 days, 10 days, 14 days, cut the tail blood of 20 μl, the blood cell analyzed peripheral blood red blood cells(RBC), white blood cells(WBC), neutrophils(N) and platelets(PLT). Sixth days after irradiation, sterilely inoculated mice unilateral femoral bone marrow cells by granulocyte colony formation test(colony forming unit granulocyte-macrophage, CFU-GM). Results After irradiation, irradiation group and hydrogen rich water group of peripheral blood cells in 14 th day to reach the minimum value,(5.0±0.1)×10^(12)/L and(7.3±0.2)×10^(12)/L, respectively; irradiation group and hydrogen rich water group of peripheral WBC of 4th day to reach the minimum value,(4.7±0.7)×109/L and(9.4±1.2)×109/L, respectively; the two group blood neutrophils 4th days to reach a minimum value were(1.4±0.4)×109/L and(3.0±0.1)×10~9/L, respectively; the two groups of peripheral blood PLT 7th days minimum value,(200.0±27.0)×109/L and(300.0±41.0)×10~9/L, respectively. Compared with the hydrogen rich water group, the water content of peripheral blood cells in the hydrogen rich water group was smaller and the recovery time was shorter. At the 6th day, the CFU-GM in the hydrogen rich water group was(124.0±8.4)×10~5/L, while that of the irradiated group was(64.0±7.4)× 105/L, the difference was statistically significant(P<0.05). Conclusion Hydrogen rich water can reduce bone marrow suppression in mice exposed to 4 Gy X irradiation.
