清化固肾排毒方对单侧输尿管梗阻大鼠肾组织Notch1和Jagged1表达的影响
Effects of Qinghua Gushen Paidu Decoction on Notch1 and Jagged1 expressions in renal tissue of rats with unilateral ureteral obstruction

摘  要: 目的观察清化固肾排毒方对单侧输尿管梗阻(UUO)大鼠肾组织Notch1和Jagged1表达的影响,探讨其抗肾纤维化的作用机制。方法 72只SPF级SD大鼠随机分为假手术组、UUO模型组、洛丁新组及清化固肾排毒方组。除假手术组外,其余各组大鼠均采用单侧输尿管结扎手术方法(UUO)建立肾纤维化梗阻大鼠模型,术后各组分别予相应药物或0.9%Na Cl溶液灌胃,每日1次。第7、14、21 d后,各组大鼠禁食不禁水1日,处死,取UUO肾组织做病理检测,行Masson染色观察其病理变化,采用免疫组织化学染色方法和实时荧光定量PCR(RT-PCR)法检测Notch1、Jagged1蛋白及mRNA的表达情况。结果随着输尿管梗阻时间延长,肾小管间质纤维化程度逐渐加重。经Masson染色后,与假手术组比较,模型组肾纤维化指数差异有统计学意义(P<0.05);与模型组比较,清化固肾排毒方组和洛丁新组大鼠肾纤维化程度相对减轻,差异有统计学意义(P<0.05)。免疫组化和PCR结果显示,清化固肾排毒方组和洛丁新组大鼠肾组织Notch1和Jagged1表达显著低于模型组(P<0.05)。清化固肾排毒方组与洛丁新组组间比较差异无统计学意义(P>0.05)。结论清化固肾排毒方可能通过下调Notch1和Jagged1来抑制Notch信号转导通路,发挥改善肾间质纤维化和保护肾脏的作用。 Objective To observe the effects of Qinghua Gushen Paidu Decoction on the expressions of Notch1 and Jagged1 in renal tissue of rats with unilateral ureteral obstruction( UUO),and discuss its mechanism on renal fibrosis. Methods 72 SD rats of SPF grade were randomly divided into the sham operation group,UUO model group,Lotensin group and Qinghua Gushen Paidu Decoction( QHGSPD) group. Except the sham operation group,rat model of renal fibrosis obstruction was established by unilateral ureteral ligation in the other groups. After operation,the rats in each group were treated with the corresponding drugs or 0. 9% Na Cl solution by intragastric administration once a day. 7,14 and 21 days after modeling,the rats in each group were fasted but drunk water freely for one day and then sacrificed. The renal tissue of obstructed side was collected,and the pathological changes of renal tissue were observed after Masson staining microscope. The protein and mRNA expressions of Notch1 and Jagged1 were detected by immunohistochemistry and real-time PCR( RT-PCR) respectively. Results As the time of ureteral obstruction prolonged,the degree of renal interstitial fibrosis gradually aggravated. After Masson staining,there was significant difference on the index of renal fibrosis between the model group and sham operation group( P < 0. 05). Compared with the model group,the degree of renal fibrosis in the QHGSPD group and Lotensin group was relatively alleviated,with statistical difference( P < 0. 05). The results of immunohistochemistry and PCR showed that the expressions of Notch1 and Jagged1 in renal tissue of rats in the QHGSPD group and Lotensin group were significantly lower than those in the model group( P < 0. 05). There were no statistically significant differences on the expressions of Notch1 and Jagged1 between the QHGSPD group and Lotensin group( P > 0. 05).
