高效液相色谱法测定维生素胶囊中合成色素

摘  要: 目的为保证保健食品的安全性,用高效液相色谱法测定脂溶性维生素软胶囊中7种人工合成色素。方法样品经石油醚萃取、乙醇-氨水-水溶液(721)溶解超声提取后,经过WAX固相萃取柱提取净化。以甲醇和乙酸铵(0.02mol/L)为流动相,梯度洗脱法经C18色谱柱分离,保留时间和光谱图定性,外标法峰面积定量。结果该法各组分检出限为0.12~0.28mg/kg,在0.5~50.0mg/L浓度范围内呈良好的线性关系,各色素组分相关系数0.9994~0.9998。样品加标回收率为86.3%~96.8%,RSD为0.9%~4.7%。结论该方法操作简便、准确可靠,能用于保健食品脂溶性维生素软胶囊中7种合成色素的检测。 Objective For the safety of health foods,to establish a high performance liquid chromatography(HPLC)method for detecting seven kinds of synthetic pigments in fat soluble vitamin soft capsules.Methods The sample was extracted by petroleum ether and extracted by ethanol aqueous ammonia water solution(7:2:1),then extracted and purified by WAX solid-phase extraction column.Methanol and ammonium acetate solution were adopted as mobile phase for a gradient elution,by chromatographic column separation,retention time and spectral were adopted for the qualitative analysis,and peak area by external standard method for quantification analysis.Results The detection limits of the method were 0.12-0.28 mg/kg.The peak area and the concentration showed a good linear correlation when the concentrations were at the level of 0.5-50.0mg/L.The correlation coefficients were 0.9994-0.9998,the mean recoveries were 86.3%-96.8%,and the relative standard deviations(RSD)were 0.9%-4.7%.ConclusionThis method is simple,accurate and reliable,and suitable for the detection of the seven kinds of synthetic pigments in fat soluble vitamin soft capsules.
