病原微生物菌(毒)种真空冷冻干燥实验风险溯源监测分析

摘  要: 目的通过菌(毒)种真空冷冻干燥模拟实验,对实验产生微生物污染风险进行研究,为对实验操作人员的危害评估和防护措施的制定提供科学依据。方法使用沙门菌作为指示细菌,对真空冷冻干燥实验活动前后的试验关键设备和操作人员出现污染的风险评估,使用物表采样和空气采样方法对实验者,实验设备、实验物品等进行污染监测分析。结果实验后生物安全柜台面、实验者手套、防护服、实验后冻干机干燥室、冻干管等均检出沙门菌,提示存在污染情况。冻干机排气管未检出沙门菌。结论菌(毒)种真空冷冻干燥实验操作在菌液制备、冷冻干燥、封口过程会产生气溶胶污染,应注意加强实验技能的培训和个人防护装备的使用。 Objective To investigate the risk of microbial contamination in the laboratory through simulated experiment of vacuum freeze drying of infectious bacteria, and to provide scientific control measures.Methods Salmonella was used as an indicator and was processed with vacuum freeze drying technique. Surface of associated equipment and tools, personal protective equipment and air samples were collected. The contamination was analyzed before and after the experiments. Results Salmonella were identified from samples collected after the experiment. These included surface of biological safety cabinet,gloves, protective clothing and the chamber and tube of freeze dryer. Samples collected from exhaust pipes of the freeze dryer were all negative.Conclusion Aerosol pollution occurred during process of suspension preparation, freeze-drying and sealing, which emphasized the importance of experimental skill training and the application of personal protective equipment.
