摘 要: 目的研究TPX22基因对乳腺癌病理进程和放疗的影响及作用机制。方法建立乳腺肿瘤干细胞裸鼠移植瘤模型,根据是否接受细胞放射干预分为干预组和未干预组,采用免疫组化法检测两组乳腺癌细胞裸鼠模型接受放疗干预前后TPX22基因蛋白表达水平,并检测TPX2相关靶基因(Bcl-2、Bax、p21及p53)在乳腺癌组织中的蛋白表达,分析TPX2与其相关靶基因蛋白的相关性。结果两组乳腺肿瘤裸鼠模型右腋下均有显著移植瘤生长,肿瘤组织与周围正常组织分解清晰,移植瘤组织内有大量肿瘤细胞弥漫性分布,肿瘤细胞形态不规则。干预组接受放疗后TPX2阳性率50%,对照组87.5%。TPX2与Bcl-2表达水平呈显著正相关性(RBcl-2=0.9688),与Bax、p21及p53呈显著负相关性(RBax=-0.9675、Rp21=-0.9563、RP53=-0.9742)。结论 TPX2在乳腺癌细胞中呈高表达,可能通过调控Bcl-2、Bax、p21及p53等相关靶基因表达水平,参与疾病进展,影响放疗效果,TPX2有望成为乳腺癌治疗的新靶点。 Objective To study the effects and mechanism of TPX22 gene in the pathological process and radiotherapy of breast cancer.Methods The nude-mouse transplanted tumor models of breast tumor stem cells were established.According to whether or not to accept cell radiation intervention, they were divided into intervention group and non-intervention group.The ex- pression levels of TPX22 gene protein in both groups were detected by immunohistochemical method, and the expression of TPX2 related target genes (Bcl-2, Bax, p21 and p53)in breast cancer tissues was detected.The correlation between TPX2 and related tar- get genes was analyzed.Results There was significant growth of transplantation tumor under right axilla of the two groups,and the boundaries between tumor tissues and normal tissues were clear.There were a large number of tumor cells which were in diffuse dis- tribution,with irregular shape.Mter radiotherapy,the positive rate of TPX2 in the intervention group was 50% while in the non- in- tervention group was 87.5%.TPX2 was significantly positively correlated with expression level of Bcl-2 (RBcl-2=0.9688),while it was significantly negatively correlated with Bax, p21 and p53 (RBax=-0.9675, Rp21 =-0.9563, RP53=-0.9742).Conclusion TPX2 is highly expressed in breast cancer cells.It may be involved in the progression of the disease by modulating the expression of Bcl- 2,Bax,p21 ,p53 and other related target genes,affecting the effect of radiotherapy.TPX2 is expected to become a new target for treatment of breast cancer.
